Study On The Brain-intestinal Interaction Of Rats With Liver Stagnation And Functional Dyspepsia Combined With Acupuncture And Medicine

Purpose:Taking Liver Depressive Type functional dyspepsia rats as the experimental object,by observing the effect of electroacupuncture,anti-depressant and electroacupuncture combined with medicine on body weight,behavior content,gastrointestinal motility,5-HT content of hypothalamus and serum,neuron specific enolase(NSE)content of hippocampus and serum of FD rats.to analyze the possible mechanism of electroacupuncture combined with medicine treatment of functional dyspepsia from zhe way of brain gut axis.Material and method:All Wister healthy SPF rats were randomly divided into 5 groups,normal group,model group,electroacupuncture group,anti-depressant,electroacupuncture combined with medicine group,10 rats per group.In the normal group,10 rats in normal feeding and away from the model environment,and avoid the environmental interference,40 rats adopted the modified chronic unpredictable mild stress model to establish the Liver Depressive Type FD model,21 days.In the treatment stage,the normal group and model group rats received daily binding stimulus,without any other treatment,for 14 days;Electroacupuncture group rats were given the bondage effect of the "Siguan"(bilateral Hegu and Tai Chong),once,15 min each,for 14 days;The anti-depressant group was treated with fluoxetine for 14 days.Electroacupuncture combined with medicine group were given the bondage effect of the "Siguan" combined with fluoxetine solution by gavage once a day for 14 days.The body weight,sugar water consumption rate,open-field test score and behavior change of each group were measured 1 day and 21 days and 35 days respectively.After the end of the experiment,the gastric emptying rate and the relative index of small intestine were measured.The levels of 5-HT in hypothalamus and serum,and NSE in serum of rats were detected by enzyme-linked immunosorbent assay(ELISA).The expression of NSE in hippocampus of rats was detected by immunohistochemistry.Results: 1.There was no significant difference in the body weight and behavior of rats in each group for the first 1 days before the experiment,and the difference was not statistically significant(P>0.05).After the modeling,the rats in the normal group and the general state before making model is basically the same,and other groups of rats showed dry hair disorder yellow,reduced activity,get together even tired lying down,stool pond,etc.;a slow increase of body weight;open field test scores of horizontal movement and vertical movement decreased significantly the sucrose preference rate decreased significantly.Compared with the normal group,the difference was significant(P<0.05).2.After the end of treatment,the intervention group weight,open field test scores are significantly increased compared with the model group(P<0.05),the total horizontal score and vertical score of the electroacupuncture combined with medicine group were significantly higher than the model group(p < 0.05);only the electroacupuncture combined with medicine group the sucrose preference rate was significantly improved compared with the model group,the electroacupuncture group and anti-depressant group(P<0.05).3.Compared with the normal group,model group rats gastric residual rate increased significantly,the intestinal propulsion rate decreased significantly,the differences were statistically significant(P<0.05);after the intervention,the intervention group rats compared with model group,reduce the rate of intestinal propulsive rate increased obviously in the stomach residue(P<0.05);the intervention groups,the gastric remnant rate had no significant difference(P>0.05),and intestinal propulsive rate difference was statistically significant(P<0.05),By comparison,the intestinal propulsive rate of electroacupuncture combined with medicine group increased obviously than anti-depressant group and electroacupuncture group(P<0.05).4.CUMS modeling can significantly decrease the 5-HT content in hypothalamic(P<0.05),after anti-depressant,electroacupuncture,electroacupuncture combined with medicine intervention,the total content of 5-ht in hypothalamus of rats increased,and anti-depressant was better than electroacupuncture group(p < 0.05),electroacupuncture combined with medicine effect was the best(p < 0.05).And the total content of 5-HT in serum,model group was significantly higher than that of the normal group(p < 0.05),electroacupuncture,anti-depressant and electroacupuncture combined with medicine intervention can reduce the serum 5-HT content(p < 0.05),the effect of electroacupuncture combined with medicine group curative effect is obviously better than that of electroacupuncture group and Western medicine group(P <0.05).the difference between electroacupuncture and anti-depressant group have no significance(P>0.05).5..CUMS modeling can significantly decreased NSE protein expression in rat hippocampus,and reduce the content of NSE in serum(P<0.05);both can enhance the expression of NSE protein in hippocampus of electro acupuncture and Western medicine,that acupuncture combined with medicine had the best effect.And the intervention group can reduce the serum total NSE levels,no significant difference between groups(P>0.05).Conclusion: 1.To establish a rat model of Liverdepression type FD,by weight,behavioral experiments and gastrointestinal motility observation,found that electroacupuncture and electroacupuncture at "Siguan" combined with fluoxetine can significantly improve the mental and physical symptoms of Liverdepressive type FD,and the curative effect of combined electroacupuncture and medicine in some respects better,proved that the electroacupuncture at "Siguan" can improve the symptoms of Liver Depressive Type FD,and can be used as a auxiliary means of drug therapy and improve the curative effect.2.CUMS modeling,he hypothalamus and serum 5-HT content,hippocampus and serum NSE content were changed in rats model of Liverdepressive type FD,compared with normal group,the content of 5-HT in the model group increased,the expression of NSE protein decreased,the total content of NSE in serum increased.Electroacupuncture and electroacupuncture combined with medicine have a benign regulation of 5-HT and NSE,proved that the mechanism of electroacupuncture and electroacupuncture combined with medicine to improve the symptoms of Liver Depressive Type FD may be to improve the activity of 5-HT and restore neuron injury.
