doses for 52 weeks,The target organ of the toxic response was observed after 13 weeks,26 weeks,39 weeks,52 weeks,and 26 weeks of administration,after 4 weeks of withdrawal.To elucidate the existence of long-term consumption of Cassia there are security risks..Methods 480 SD rats were randomly divided into 4 groups according to body weight.The rats in the control group(CN group),low dose group(T1 group),middle dose group(T2 group),high dose group(T3 group),120 rats in each group,Male and female half.The difference in body weight between the two groups did not exceed 20%of the mean body weight.Administered 6 days a week,once daily dosing,continuous administration of 52 weeks,oral(IG)administration,administration Volume:1.0ml/100g of body weight.Administration dose are:0.5g crude drug/KgBW,2.2g crude drug/KgBW,10.0g crude drug/KgBW.Respectively,equivalent to the amount of human body equivalent dose of 0.5,8.8 and 40.0 times(human weight according to 60Kg calculation).The control group was given equal volume of purified water.Animals were sacrificed 20/group,male and female half at the 13th,26th,39th and 52th week and 26,52 weeks after the administration of the drug for 4 weeks.Intraperitoneal injection of pentobarbital anesthesia,Hematology and serum biochemical measurements were performed from the celiac arteries.The weight of the organ was measured and the organ coefficient was calculated.The organism was examined by all the organs in each dose group.Results 1.the whole test cycle did not occur due to given cassia seed drug itself caused by dying and death of the rat toxicity.2.The dose group and the control group were increased body weight,showing a dose-response relationship.3.cassia rats administered intragastrically 26 weeks,total bilirubin dose,high dose group and the high dose female rats in groups of male rats(TBIL)lower(P<0.01),after 4 weeks of recovery,high anf the total bilirubin(TBIL)in the male rats was still low(P<0.01).After administration for 39 weeks,the total bilirubin(TBIL)was low in the high dose group(P<0.05 or P<0.01).After administration for 52 weeks,the triglyceride(TG)in the high dose group was significantly lower than that in the control group(P<0.05).After four weeks of recovery,triglyceride(TG)returned to normal.4.Rats were given intragastric administration of Cassia 26 and 39 weeks,the rats in the middle dose and high dose group were significantly higher than those in the control group(P<0.05 or P<0.01).5.After 13 weeks of continuous injection of Cassia obtusipiro,SD rats did not show toxicological changes related to the samples.After continuous administration of Cassia 26,39 and 52 weeks,the target organism in the high dose group was kidney and renal toxicity injury is irreversible,in the low dose group did not appear obvious toxicity.Conclusion The no-observed-adverse-effect level for Semen Cassiae freeze-dried powder was 2.2 g/kgBW in rats(Equivalent to 8.8 times the amount for human),and the toxic target organ in high-dose 10.0 g/kgBW(Equivalent to 40.0 times the amount for human)group was kidney.Background allergic contact dermatitis(ACD)is a late-type globally,is one of the important chemical substances induced by ACD,and further elucidates the pathogenesis of nickel allergic contact dermatitis as an important problem that needs to be solved urgently.X-ray fluorescence analysis(XRF analysis)is an advanced lossless element analysis method,synchrotron radiation is XRF analysis of the ideal X-ray light source.In this study,we established guinea pig and mouse nickel allergic contact dermatitis animal model,and the use of synchrotron radiation X-ray fluorescence analysis of nickel allergic contact dermatitis pathogenesis to explore.Objective 1.To establish the animal model of allergic contact dermatitis;2.To reveal the distribution of nickel in skin tissues under the pathological state of nickel ACD;3.To reveal the distribution of copper,iron,zinc,sodium,potassium,calcium,magnesium under the pathological state of nickel ACD.Methods:1.the establishment of nickel allergic contact dermatitis guinea pig and mouse model;2.access to a representative of the nickel allergic contact dermatitis guinea pigs and mouse skin lesions specimens,after a specific treatment by synchrotron radiation X The content of nickel in the depth of lesion was analyzed by X-ray fluorescence analysis,and the data of nickel were obtained by the same detection method in guinea pig and mouse skin samples.Results Nickel allergic contact dermatitis guinea pigs and nickel allergic contact dermatitis mice were successfully obtained by sensitization and stimulation of nickel sulfate at different time points.Through the analysis of guinea pig skin samples and mouse skin samples,it is found that nickel can enter the body through the epidermis and accumulate in the skin tissue,which gradually diminishes with time;the content of nickel in the skin at each depth is affected by the individual With or without nickel allergic contact dermatitis,individual nickel allergic dermatitis showed acute or chronic,where the skin contact with nickel concentration,sensitization time and showing a different distribution of micro-area.Conclusion:1.The animal model of allergic contact dermatitis can be successfully established by the sensitization and excitation of nickel sulfate.(2)The distribution of nickel in the skin tissue has a series of characteristics under the pathological condition of nickel allergic contact dermatitis;3,synchrotron
