Objective:Sophorae Tonkinensis Radix et Rhizoma—as a clinical commonly used medicine which has been reported repeatedly that it leads to poisoning in clinical use.One of the reasons is that Sophorae Tonkinensis Radix et Rhizoma is always confused with Menispermi Rhizoma.At present,there is a lack of systemic research about the quality standard of Sophorae Tonkinensis Radix et Rhizoma.This research can provid a reference basis and train of thoμght for the quality standard of Sophorae Tonkinensis Radix et Rhizoma by finger-print,quantitative determination and component analysis which is conducted by high resolution mass spectrum(UPLC-Q-TOF/MS).Methods:(1)UPLC-PDA was used in finger-print of the 70%methanol extracts of Sophorae Tonkinensis Radix et Rhizoma.The data was analyzed with a soft "Smilarity Evaluation System for Chromatographic Fingerprint of TCM" published by GPC(Version 2004A)and the common model of chromatographic fingerprints,common peaks and similarity were calculated.Then oxymatrine was specified as reference peaks,the relative retention time and relative peak area were calculated to evaluate the difference of Sophorae Tonkinensis Radix et Rhizoma from different habitats.(2)This research confirmed the best extraction condition(including extract method,extract solvent and extract time)and HPLC-PDA to confirm the best chromatographic condition by using single-factor test.Then the quantitative determination of matrine,oxymatrine,sophocarpine and oxysophocarpine could be implemented.The raw data was analyzed by using SAS 8.2 to finish cluster analysis and the habitats were divided into three categories.(3)UPLC-Q-TOF/MS was used to detect the compositions of Sophorae Tonkinensis Radix et Rhizoma.The raw data could be analyzed by the software Unify and the potential compositions as well as the fragmentation patterns could be confirmed.Results:(1)The 27 common peaks were confirmed and 2 common peaks were specified as matrine,oxymatrine;the relative retention time’s RSD of common peaks from different habitats is between 3.925%-4.588%,but the relative peak area’s RSD is large.The similarity is greater than 0.9 of most habitats and the similarity of Menispermi Rhizoma.is poor.(2)The content of matrine and oxymatrine could reach pharmacopoeia standard(≥0.7%)except one habitat.The habitats were divided into three categories according to the content of four alkaloids(including matrine,oxymatrine,sophocarpine and oxysophocarpine).(3)The final result was inferred for 56 components according to the matching results of Unify.Conclusion:(1)In this research,Sophorae Tonkinensis Radix et Rhizoma from different habitats has good similarity and the method which has good precision,stability,repeatability,characteristic and strong specificity could provide new ideas for the identification of Sophorae Tonkinensis Radix et Rhizoma and Menispermi Rhizoma.So that to avoid of drug poisoning caused by a mixture of these tow kinds of herbs.(2)The results showed that there was a big difference of contents of four alkaloids between different habitats.But the contents in geo-authentic habitat was relatively high.Most areas where’s contents were greater than 1.0%are in Guangxi province,it was in accordance with the fact that Guangxi province is the geo-authentic habitat.The content of matrine was inversely proportion to the oxymatrine’s.The result of cluster analysis showed that habitat has a great influence on the contents of alkaloids,difference between different groups is so small.The result reminded that Sophorae Tonkinensis Radix et Rhizoma from more habitas should be collected and analyzed.(3)The results provide a reference idea for further chemical research of Sophorae Tonkinensis Radix et Rhizoma.But the inference results require further validation.
