Objective:Abri cantoniensis Herba is the dried whole plant of Abrus cantoniensis Hance,which is mainly distributed in Kwangtung and Kwangsi provinces of China and this plant is a native herb there.This herb is considered to be medicinal for the whole plant and have medicinal properties for clearing heat and eliminating damp,dispersing the depressed liver-qi,alleviating pain and promoting blood circulation to remove blood stasis etc.Modern pharmacological research show that the Abri cantoniensis Herba has functions of protecting liver,antioxidant,antiinflammation,antibacterial,antivirus,immunomodulatory,lipid-lowering and other pharmacological effects.In clinical practice,the herb is commonly used to treat liver disease.By consulting literature we can learn that Abri cantoniensis Herba contains flavonoids,triterpene,alkaloid,anthraquinone,polysaccharide,organic acids,volatile oil and other chemicals.Among them the alkaloids,flavonoids,triterpene are the main active ingredients.But so far the research of Abri cantoniensis Herba on quality control is mainly focused on the determination of the content of alkaloids,and the research on flavonoids is less,not to mention the attention on triterpene.At the present stage,combining fingerprint with multi-component content determination is a kind of quality control mode which is more suitable for Chinese traditional medicine.Because it conforms to the fuzzy characteristics of Chinese medicine,and makes Chinese medicine quality control more scientific and comprehensive.In Chinese Pharmacopoeia(2015),Abri cantoniensis Herba Only has qualitative identification and has no content determination item,which is no good to the quality control of the herb.Therefore it is very necessary to establish a comprehensive,reasonable quality standard,providing the basis for the improvement of the quality standards for Abri cantoniensis Herba.Method:(1)The UPLC fingerprint of Abrus cantoniensis Hance was established,and similarity analysis,hierarchical clustering analysis,and principal component analysis were undertaken to study 16 sets of UPLC fingerprints of Abrus cantoniensis Hance.(2)Adopting UPLC/Q-TOF-MSE technologies combined with database of UNIFI natural products and database of Abrus cantoniensis Hance to analysis the chemical composition of Abrus cantoniensis Hance rapidly.(3)To establish an UPLC method for simultaneous determination of abrine,hypaphorine,vicenin-2,schaftoside,and determining contents of 16 sets of medicinal materials to provide a method for improvement the medicinal materials quality standard.(4)To establish a HPLC-DAD-CAD fingerprint method of Abrus cantoniensis Hance,Similarity analysis was undertaken to study 16 sets of UPLC fingerprints of Abrus cantoniensis Hance.(5)Using the zebrafish liver injury model induced by thioacetamide to do preliminary study on the protective effect of Abrus cantoniensis Hance for liver injury.Results:(1)The UPLC fingerprint of Abrus cantoniensis Hance was established and 52 peaks were selected as the common peaks,identify the 7 common peaks.The similarity analysis,hierarchical clustering analysis,and principle component analysis were employed for the quality evaluation of 16 batches of samples,three results are mutually authenticated to enhance the credibility of the results.The results showed that 16 species of Abrus cantoniensis Hance were divided into four classes,Guangdong Zhaoqing samples were the wild as a class,two batches of Guangdong cultivated herbs as a class were different from Guangxi samples,it can be seen that the quality of samples from Guangxi and Guangdong is different;and one of Guangxi Qinzhou samples were classified alone as a class,showing that even the same origin,there are individual differences in the quality.(2)To establish an UPLC method for content determination of four effective components.Methodology investigation show that the method repeatability,stability,precision and recovery tests are in accordance with Chinese Pharmacopoeia,can be used as a valid content determination method.(3)The fingerprints of HPLC-DAD-CAD were established for the first time.By comparing the DAD and CAD fingerprints,we found that some of the less polar components were less or no response under the UV detector.Mass spectrometry data analysis and identification,we can see that Abrus cantoniensis Hance contains many triterpenoid saponins,in the CAD detector has a good response,is one of the main components.The necessity and importance of the introduction of CAD detector provides the ideas and methods for the further study of triterpenoids in Abrus cantoniensis Hance,and strive to evaluate the quality.(4)The database of Abrus cantoniensis Hance was established.The data were analyzed and matched with UNIFI,and then were tested by artificial check.A total of 40 compounds were identified,and the possible pyrolysis of flavonoids and triterpenoid saponins was speculated.(5)Verify the zebrafish liver injury model induced by thioacetamide,and use the model to do preliminary study on the protective effect of Abrus cantoniensis Hance for liver injury.The results show that Abrus cantoniensis Hance has some protective effect on liver injury induced by thioacetamide.Conclusion:This study established a UPLC fingerprint combined with four kinds of methods for the determination of active ingredient contents of Abrus cantoniensis Hance,HPLC-DAD-CAD fingerprint,providing the basis for the quality control and the improvement of the quality standards for Abri cantoniensis Herba;Adopting UPLC/Q-TOF-MSE technologies to analysis the chemical composition of Abrus cantoniensis Hance,which can help the research on the pharmacodynamic material basis and provide the quality control of material basis and data support,and also is conducive to further research and development of the herb;Studies have shown that Abrus cantoniensis Hance on liver injury induced by thioacetamide has certain protective effect,and the existing research results verify each other.It provides a new way of thinking for the screening and research on other protect liver medicine.
