Objective:To investigate effects of QingreJieDuHuaZhuoPian on rat models of pancreatic and liver tissue morphology and liver function(ALT,AST,TBIL,DBIL,ALB,GLB,A/G),endotoxin,PECAM-1,IL-1 and IL-4,that pancreatic injury of rats is induced by intestinal endotoxemia,witch is induced by alcohol and high fat.To investigate whether the mechanism of prevention and cure is related to the reduction of inflammatory reaction and the protection of pancreatic microcirculation.Methods:70 SD rats were randomly divided into normal control group(n = 10)and model control group(n = 60).The normal control group was fed with distilled water and ordinary feed.The model group was treated with alcohol and high fat diet.The model of IETM pancreatic injury was established in rats.At the end of 3 monthes,4 odel rats were randomly selected to determine the success of modeling,the remaining 56 rats were randomly divided into qinhua group and model control group with 28 rats in each group.The model control group was fed with distilled water and ordinary diet,qinghua group was fed with QingReJieDuhuaZhuoPian suspension and normal diet for 4 weeks,respectively.The liver function and endotoxin were detected by abdominal aorta respectively.The expression of PECAM-1,IL-1 and IL-4 in pancreas were observed by immunohistochemical method.The pathological changes of liver and pancreas were observed by HE staining.Result:1.The pathological morphology of pancreatic tissue in rats: model control group of pancreatic acinar hypertelorism with acinar structure damage,inflammatory cell infiltration,focal necrosis,pancreatic lobules isolated from necrotic areas,red meat and transparent thrombosis.Normal control group had normal pancreatic tissue,no obvious degeneration and inflammatory cell infiltration.qinghua group than in the model control group reduces the width of acinar spacing increases,there is little inflammatory cell infiltration.2.The pathological morphology of rat liver: The model group showed liver cell morphology of rat liver cells appeared irregular,a large number of fat cells to push the product,fatty degeneration of liver cells increased,hanging in the central nucleus,or be crowded with one side,liver sinus extrusion narrowing,liver cords disorders,inflammatory cells with varying degrees of infiltration,spotty necrosis,even there were few fibrous degeneration.Qinghua group rats hepatocyte morphology still rules,hepatic cord arranged neatly,liver cells can be seen between a few lipid droplets filled between the liver cells,but no fusion of lipid droplets,no significant inflammatory cell infiltration and necrosis.3.Endotoxins: Pasma endotoxin levels of model control group were higher than the normal control group and Qinghua group,and were highest.The differences compared with the normal control group had statistical significance(P<0.01),compared with qinhua group had significance(P<0.05).4.Liver function: The model control group ALT,AST,GLOB,GGT compared with the normal control group and qinghua group increased(P<0.01).After the treatment,ALT,AST and GGT,of qinghua group were significantly decreased(P<0.01).The ALP,DBIL and IBIL in model group were higher than those in normal control group(P<0.05),ALP and DBIL were significantly decreased after treatment(P<0.05).The ALB of model control group had no statistical significance(P>0.05),compared with normal control group and qinghua group.5.IL-1 and IL-4: The expression level of IL-1 in the model group was higher than that in the normal control group and qinghua Group.The differences were statistically significant(P<0.01),that the model group compared with the normal control group,and were significant,that compared with qinghua group(P<0.05).The expression of IL-4 in pancreas of qinghua group were higher than the normal control group and model group.There was significant difference between the control group and the model group(P<0.05);compared with normal control group,the expression of IL-4 n pancreas of model control group was increased,the difference was statistically significant(P<0.05).6.PECAM-1: The expression level of PECAM-1 in pancreas of model control group was higher than that of normal control group Group and Qinghua Group(P<0.01).Conclusion:The mechanism of action tha Prevention and treatment of intestinal endotoxemia induced by alcohol and high fat in QingReJieDuHuaZhuoPian may be reducing or eliminating endotoxin,promoting the expression of anti-inflammatory cytokine IL-4,thereby inhibiting the expression of proinflammatory cytokines IL-1,coordinating dynamic balance between proinflammatory and anti-inflammatory cytokines,improving liver function,reducing liver injury,reducing systemic inflammatory response and reducing PECAM-1 level,So to protect the microcirculation of pancreas and pancreatic injury. |
