Objective:To observe the effect of Qi-Shao Jiang-ya compounded tablets on Spontaneously hypertensive rats(SHR) blood pressure, serum levels of IL-1β and IL-10. Try to explain the mechanism of antihypertensive and protect myocardium from the angle of interventing the inflamematory of hypertension, provides a theoretical basis for the Qi-Shao Jiang-ya compounded tablets in treating patients with hypertension.Methods:Animal grouping:50 SHRs (8 weeks old male) were randomly divided into 5 groups:Spontaneously hypertensive rats model group (SHR Group), Irbesartan group(Irbesartan Group), Qi-Shao Jiang-ya compounded tablets high dose group (Qi-Shao Jiang-ya high dose group), Qi-Shao Jiang-ya compounded tablets middle dose group (Qi-Shao Jiang-ya middle dose Group), Qi-Shao Jiang-ya compounded tablets middle dose group (Qi-Shao Jiang-ya low dose Group).Another 10 WKY rats into blank control group (WKY Group). After 8 weeks treatment, ELISA method for the determination of serum IL-1β,IL-10 levels, the HE staining to observe the myocardial tissue morphology.Result:(1) The effect of SHR pressure:Each group of SHR’s blood pressure equivalent before gavage treatment (P> 0.05), significantly higher than that in WKY group (P<0.05), suggesting that hypertension model is reliable.There was no significant change in blood pressure between SHR group and WKY group, no statistical significance (P> 0.05). The blood pressure of experimental group (Irbesartan Group, High dose group, Middle dose Group, Low dose Group) were decreased significantly compared with before treatment (P<0.05).In Irbesartan group, High dose, Middle dose group, the three groups decreased significantly and lower than the Low dose group of blood pressure, the difference was statistically significant (P<0.05).(2)Effect of serum IL-1βp and IL-10 levels in rats:The level of serum IL-1β in model group was significantly higher than that in the WKY group, the difference was statistically significant (P< 0.05). The serum IL-10 level in the SHR group was significantly lower than that in the WKY group, and the difference was statistically significant (P< 0.05).The influence on the concentration of serum IL-1β:Experimental group (Irbesartan group, High dose group, Middle dose group, low dose group) serum levels of IL-1β were lower than that of SHR group (P< 0.05), and higher than the WKY group (P< 0.05). The serum level of IL-1β in the Middle dose group was lower than that in Irbesartan group and Low dose group (P< 0.05), and equivalent to that of the high dose group (P> 0.05).(3) Effects on the morphology of cardiac muscle in rats:In the SHR group and Irbesartan group, the myocardial fibers were arranged in disorder, the cell nucleus became smaller and smaller, some of the cells were dissolved, and the nucleus disappeared. In the High and Middle dose group group, the cardiac muscle fibers were arranged in a regular, cardiac muscle cell hypertrophy, and the nucleus deviated from the center of the cell. Compound Qishao low group of rat myocardial cells in lipid droplets, cell hypertrophy, obviously karyopyknosis becomes small. In the WKY group, the myocardial fibers were in the regular pattern, and the cardiac muscle cells arranged regularly, and the nucleus was located in center.Conclusion:(1) Qi-Shao Jiang-ya compounded tablets can effectively lower blood pressure and protect myocardium;(2) Qi-Shao Jiang-ya compounded tablets lowering blood pressure and protect myocardium mechanisms may be:By inhibiting the production of pro-inflammatory factors, and promoting the formation of anti-inflammatory factors, regulate the inflammatory balance, and thus play a role in reducing blood pressure. |
