Objective:Using two-dimensional gel electrophoresis and mass spectrometry of blood heat,blood stasis type, blood for serum and normal serum protein spectrum prepared using the protein-protein interaction between different syndromes compare normal human serum and serum samples differences in protein composition to explore parts of psoriasis different syndromes unique expression of the protein, revealing the syndrome and psoriasis associated with the formation of specific proteins and their biological characteristics relationship.Methods:Select the Second Affiliated Hospital of Hunan University of Traditional Chinese Medicine Dermatology in October 2013- December 2014 among patients with psoriasis vulgaris patient department treated 90 cases as the observation group(blood stasis syndrome in 30 cases, 30 cases of blood deficiency, blood heat syndrome 30 cases); select concurrent examination of 30 cases of healthy people as a control group, two groups were collected serum object of study, and protein extraction and quantification of two-dimensional electrophoresis, etc., and finally the use of mass spectrometry and database searching to identify two differentially expressed proteins in serum, and psoriasis different syndromes of different proteins.Results:According to two-dimensional electrophoresis and mass spectrometry results were successfully identified 20 kinds of proteins, psoriasis group compared with the control group,patients with psoriasis blood fibrinogen β chain, blood fibronectin, fibrinogen γ chain, HPX protein, α- antitrypsin, apolipoprotein A-ⅳ, complement C3, lectin(apolipoprotein,fragments), serum transferrin, between-α- trypsin inhibitor heavy chain H4(fragment),apolipoprotein E, complement C4-B, mannose-binding protein-C, leucine-rich-α-2- albumin and serum glycoprotein expression was significantly increased, while serum albumin(fragment), transthyretin, IgαC chain contained lipoprotein L1, binding globin expression is significantly reduced. Wherein the serum of psoriasis patients have abnormally high expression of partial protein, while the expression of a certain difference with psoriasis different syndromes, complement C3, lectin, serum transferrin protein deficiency in psoriasis the patient’s serum was higher than the other two syndromes of psoriasis, and the expression of β chain of fibrinogen, fibronectin and other blood expression levels in serum of patients with psoriasis blood heat and blood stasis is higher than psoriasis patients with.Conclusion:Different syndrome types and healthy people with psoriasis vulgaris group protein expression patterns exist obvious differences, the analysis found that the main reason for the difference in different related to the pathogenesis of possible and different syndrome types. |
