| Objective: We explored the possible mechanism and provide theoretical foundation of electroacupuncture prevention and treatment on postmenopausal osteoporosis by observing bone mineral density and LP and OB-R in hypothalamic in ovariectomized rats after electroacupuncture treatment.METHOD: Forty three-months healthy female Sprague-Dawley rats were randomized into a sham operation group,a model group,a medication group and an EA group,10 rats in each one.Samples in the sham operation group removed the adipose tissue near the ovaries,the other three groups removed the ovaries,adaptability of rats fed for five days after surgery. EA group were applied to “Shenshu”, “Zusanli”, “Guanyuan”, “Sanyinjiao” and drenched saline, medication group were treated with Alendronate and given fixed, sham operation group and model group were drenched saline and given fixed, treatments were both given once a day,ten times a course of treatment,and totally nine courses were given.Separated the hypothalamic and right femur of rats after the end of the course, the bone mineral density were detected by dual-energy X-ray absorptiometry, LP m RNA and OB-R m RNA were detected by RT-PCR.Results: 1 The bone mineral density of rats reduced after ovariectomized,the BMD of model group, medication group and EA group were lower than sham operation group,but the BMD of EA group and medication group higher than model group. 2 The LP and OB-R in hypothalamic of model group were higher than sham operation group, the LP and OB-R in hypothalamic of EA group and medication group were lower than model group. Conclusions: 1 The electroacupuncture treatment on Shenshu, Zusanli, Guanyuan, Sanyinjiao was able to slow down the bone loss of ovariectomized rats,was able to prevent postmenopausal osteoporosis. 2 The electroacupuncture treatment on Shenshu, Zusanli, Guanyuan, Sanyinjiao can cause the inhibition of the raise of the LP and OB-R in hypothalamic of ovariectomized rats,this adjusting way possibility had the effect that prevent bone loss after menopause. |
