Nowadays,knee osteoarthritis of knee osteoarthritis(KOA)has been developed for clinical common and frequently occurring disease,and with the development and progress of acupotomology,knife in the clinical curative effect is distinct,needs to have a related research to provide theoretical support.This thesis is from the surrounding soft tissue as a starting point for experimental study,to further clarify the relevant mechanical mechanism of acupotomy in the treatment of knee osteoarthritis model rabbits,to further expand the clinical treatment of KOA ideas and methods.ObjectiveThe experimental observation of acupotomy therapy on osteoarthritis of knee joint osteoarthritis model rabbit medial collateral ligament mechanical changes and interleukin 4 mediated chondrocyte mechanics signal transduction efficacy of intervention,of acupotomy intervention KOA mechanics and repair of cartilage cell mechanism.MethodsThe six month old healthy New Zealand rabbits 24(male and female)and were randomly assigned to four groups(normal group,model group,electro acupuncture group,Acupotomy group),each with 6 rabbits in each by left hindlimb extension fixation method of preparation of knee osteoarthritis model.After 6 weeks of braking,the KOA model rabbit was evaluated by X-ray image,and the model was successful or not..After modeling success 1 week after treatment,a total of 3 weeks of treatment,to normal feeding for 3 weeks,drawing,on four groups of rabbit koa were knee medial collateral ligament mechanical testing(force experimental relaxation,creep test,tensile test),analysis of KOA model rabbit medial collateral ligament in mechanical changes before and after modeling and therapeutic intervention after;and at the same time,the content of IL-4 ELISA quantitative detection of KOA model in rabbit chondrocytes,analyses before and after modeling and after therapeutic intervention after chondrocytes cultured in IL-4 content changes.ResultsThe results of medial collateral ligament of knee joint mechanics test:1 The tensile test results show that:After the success of the joint brake model,compared with the normal group,the model group had a significant difference in the tensile mechanical properties of the medial collateral ligament of the knee joint.Model group MCL limit load,the maximum should force and maximum strain were significantly decreased,the ultimate load and the maximum strain and the normal group in a statistically significant difference(P<0.05),and the maximum stress and normal group compared with significant difference(P<0.01).Acupotomy group intervention for 3 weeks after the electro acupuncture intervention after 3 weeks,compared with the model group,Acupotomy group limit load,maximum stress,maximum strain were increased;Electroacupuncture group limit load and the maximum strain is increased,the maximum stress did not change.Three the tensile index of acupotomy group and electro acupuncture group is high.That has certain effect on tensile properties of ligament acupotome can rabbit model,and acupuncture group than in the electroacupuncture group.2 The creep experimental results show:Joint immobilization made successful model,in the model group,the creep rate than that of the normal group significantly increased,after Acupotomy group intervention for 3 weeks and electro acupuncture after 3 weeks,compared with the model group,Acupotomy group and electro acupuncture group of creep rate have obvious decreasing trend,but statistical differences between the four groups is not obvious(P>0.05).3 The stress relaxation experiment results show:The stress relaxation rate of the ligament in the model group was significantly higher than that in the normal group after the joint brake model was successful..Acupotomy group intervention for 3 weeks after the electro acupuncture intervention after 3 weeks,compared with the model group,Acupotomy of model group ligament relaxation rate produced a good influence,Acupotomy group and electro acupuncture group should stress relaxation rate decreases,and acupotomy group decreases more than electro acupuncture group,acupuncture and electroacupuncture can change stress in good model of the stress relaxation rate of ligament stress relaxation characteristics of different degrees of impact,but statistical differences between the four groups is not obvious(P>0.05).4 IL-4 in chondrocyte test results show that:After the joint immobilization,the content of IL-4 in the model group was significantly decreased,and the difference was significant(P<0.01).Acupotomy group intervention for 3 weeks after the electro acupuncture intervention after 3 weeks,compared with the model group,Acupotomy group and electro acupuncture group,the cartilage cells of IL-4 content increased,Acupotomy group almost to the normal group,Acupotomy group has difference(P<0.05)and EA group no difference(P>0.05).Compared with the normal group,Acupotomy group no difference(P>0.05)and EA group has difference(P<0.05).Show that acupuncture and electroacupuncture intervention can increase the content of IL-4 in cartilage and acupotomy intervention has certain advantages,articular cartilage injury repair and improve the role.Conclusion1 Acupotomy intervention can adjust the biomechanical properties of the ligaments,is conducive to the recovery of knee joint mechanical balance..2 Acupotomy intervention of interleukin-4 mediated chondrocyte mechanics signal transduction has a positive regulatory role,and can promote the repair of articular cartilage of the knee.
