Three new organic small molecular fluorescent probes have designed and synthesized,based on the fluorophores of Green fluorescent protein—HOBDI and BODIPY.Theses probes exhibited the properties of aggregation induced enhanced emission(AIEE)and Intramolecular charge transfer(ICT),which could be used to quantitative detection in biological system.(1)A new organic small molecule fluorescent probe was designed and synthesized based on the HOBDI,one GFP similar fluorophore,and its photo physical properties were preliminarily tested.Because the probe molecules are not rigid conjugated planar structures and the carbon-carbon(C-C)bonds between nitrobenzene and imidazolidinone can rotate,the probe Lyso-3 has the characteristics of aggregation induced emission.(2)Design and synthesis of a new fluorescent Lys-V for highly selective detection of lysosomal viscosity.The viscosity increased can restrict the C-C twist around the exocyclic double bond between phenol and imidazolidinone,so the fluorescence intensity of Lys-V would enhance with the increase of viscosity and it is stable in different pH solvents at same viscosity,which insure that the probe Lys-V can be used to monitor change of viscosity in different pH condition.Therefore,the probe can be used to map viscosity in live cells because of its ideal water solubility and low cytotoxicity.And the probe can selectively target lysosomes with its weakly basic amines morpholine substituents.(3)A new type of fluorescent probe for tyrosinase was designed and synthesized based on BODIPY and coumarin through the FRET mechanism.With excitation,fluorescence of coumarin could be transferred to irradiate BODIPY and final give the fluorescence signal of BODIPY.After the probe was treated with tyrosinase,which could catalysis 4-hydroxylphenyl into quinone,and the fluorescence of BODIPY was quenched because quinone(Donor)transfer electronics to BODIPY(Acceptor).Thus,the FRET was inhibited and fluorescence of coumarin comes out.The fluorescence changes between the coumarin and BODIPY channels could be used for quantitative determination of tyrosinase by ratiometric sensing in living cells.
