The purpose of this study was to investigate the ability of peptidoglycan(PG)microspheres inducing different parts of mouse to express peptidoglycan recognition protein(PGRP)mRNA and enhancing antigenic activity.In this study,PG or PG microspheres were administered to mice through the oral route.After one week,the cytokines in serum were detected in different periods.After 21 days post gavage,quantitative PCR technique was used to detect the expression of PGRP mRNA in liver ? kidney?spleen and thymus of mice.The results showed that the content of PGRP mRNA in liver and thymus of PG microspheres group were higher than that of blank control group,and the difference was very significant(p<0.01).Meanwhile,the content of PGRP mRNA in kidney and spleen were higher than that in control group,and the difference were significant(p<0.05).The content of PGRP mRNA in four organs of PG microspheres group were higher than that of PG group,and the difference were significant in liver,spleen and thymus(p<0.05).The levels of IFN-?,TNF-?,IL-4 and IL-12 in serum of PG microspheres group were higher than those in blank control group;The content of IFN-??TNF-? and IL-12 in serum of PG microspheres group were significantly higher than that of blank control group(p<0.01)and IL-4 were significantly different from those in control group(p<0.05).In order to study the effect of PG microspheres on the antigen activity,bovine serum albumin(BSA)was used as a model antigen.After administered to mice through the oral route,the mice were injected subcutaneously with BSA.The results showed that the TNF-?,IL-4 content in serum of PG microspheres group compared with the blank control group were significantly different(p<0.01).The contents of IgG1 and IgG2 a were significantly different from those of control group;In BaLb/c mice,the IFN-,TNF-,and IL-4 in serum of the PG microspheres group were significantly different compared with the blank control group(p<0.05),the difference of IgG content was significantly different with the control group(p<0.01).This study further showed that PG microspheres can increase the expression of PGRP mRNA in the body and enhance the immune function of the body.
