Rheumatoid arthritis(rheumatoid arthritis RA)is a kind of autoimmune disease.This disease mainly induce chronic and symmetrical inflammation,resulting in synovial erosion of peripheral joints[1-3],its pathogenesis is not yet clear.Modern medicine consider that RA is a disease closely related to environment,cell,virus,heredity,sex hormone and mental state.According to the findings of the survey,in recent years rheumatoid arthritis induced disability,second only to cardiovascular disease induced disability.The early onset of 1 to 2 months as the"window period",if effective treatment,the illness will be significantly control.If in the early stage of the disease did not receive effective treatment,the disease will be protracted,until the chronicity of development.Therefore,to find the early diagnosis indicators of rheumatoid arthritis is the premise of its early treatment.According to the latest research,monocytes have a great change in the development of rheumatoid arthritis.Especially,the change of subtype is more obvious.This change is a powerful proof of the patient’s condition.so it will provide good help for early treatment.Monocyte is an important antigen presenting cell in peripheral blood and plays an important role in the development of inflammation.Our previous studies have found that the changes of CD14+CD16+monocytes in RA are obvious,so this study will be able to study whether it can be used as an indicator to indicate the changes of RA patients as early as possible.Objective:To detect the proportion of monocytes and subsets in patients with rheumatoid arthritis(RA)and to reveal its role in the pathogenesis of rheumatoid arthritis(RA).Methods:1 Peripheral anticoagulation blood was collected from 76 patients with RA(disease activity group)and 57 healthy controls(normal control group),and the proportion of CD14+monocytes and their subsets(CD14+CD16+,CD14+CD16-monocytes)was detected by flow cytometry.2 The clinical data of patients with RA were analyzed,and the correlation between monocytes and their subsets and clinical features was analyzed.The role of monocytes and their subsets in the pathogenesis of RA was preliminarily revealed.Results:1 The number of monocytes in peripheral blood of patients with RA was significantly increased(19.89+1.584 vs.14.46+1.207,P<0.05).The results of monocyte subsets showed that the proportion of CD14+CD16+subsets increased significantly(20.43+0.7306 vs.10.75+0.3822,P<0.001),while the proportion of CD14+CD16-subsets decreased(80.11+1.584 vs.85.54+1.207,P<0.05)2 The results of clinical correlation analysis showed that CD14+monocytes,especially the CD14+CD16+subgroup,were positively correlated with the condition of RA patients,and positively correlated with the clinical parameters of ESR,CRP,CCP and DAS28.Conclusion:In patients with RA,the number of monocytes increases significantly,and the change of subgroup composition(CD14+CD16+abnormal increase of monocytes)aggravates the progression of RA,so it can be used as an indicator to indicate the change of RA patients as early as possible.
