Objective Explore the mechanism of Yangjing capsules(YC)affecting testicular microcirculation in dyszoospermia mice exposed to cyclophosphamide(CP).Methods Adult mice were divided into control,model and YC treatment group with low,medium and high dose.The morphologic change of the testis were detected by paraffin section HE staining techniques.The expression of testicular CD34 was detected by immunohistochemistry of paraffin section and Western-Blotting.The m RNA and protein expression of VEGFA,VEGFR1,VEGFR2,NOS3,OCLN,AKT1,SRC and STAR were detected by RT-q PCR and Western-Blotting.Results Both dyeing depth and protein expression of CD34 in the model group were significantly lower than that in the control group,high dose YC group rised significantly to protein expression of CD34.Neither the testicular m RNA expression of VEGFA,VEGFR1,VEGFR2,NOS3,AKT1,SRC or STAR in model group mice was different compared with the control group.Only OCLN expression was significantly higher than that in the control group.The protein expression of the 8 factors above were significantly lower in the model group compared with the control group.But there were no differences among model and treated group on AKT1 protein expression.While NOS3 expression in low dose group,VEGFR1,STAR expression in medium dose group,VEGFA,VEGFR2,SRC expression were all significantly higher than those in the model group.Conclusion YC can significantly improve the testicular microcirculation in CP mice through raising angiogenesis factors(VEGFA,VEGFR1,VEGFR2,NOS3,AKT1,SRC)and Tight junction protein(OCLN).The spermatogenesis improvement of YC was closely related to its enhancement to testicular microcirculation,and the angiogenesis pathway may also affect spermatogenesis by regulating testosterone synthesis.
