| Objective To investigate whether resveratrol can prevent the release of aseptic loosening by activating the autophagy in the presence of wear particles of titanium alloy to improve osteogenic differentiation of bone marrow mesenchymal stem cells.Methods Resveratrol at different concentrations(0,0.125,0.5,1,2.5,5,and 10 μmol/L)was applied to bone marrow mesenchymal stem cells from C57 mice,Caspase-3 detects cellular activity to determine the optimal duration and concentration of the drug.Bone marrow mesenchymal stem cells(BMSCs)were treated with the best duration and concentration of drug.Autophagy markers Atg5 and Lc-3 a/b were detected by western-blot to identify autophagy of bone marrow mesenchymal stem cells.happened.Bone marrow mesenchymal stem cells were induced by osteogenic differentiation induction medium for 21 days.ALP activity in the culture medium was detected.Alizarin red staining was performed and quantitative analysis was performed by using cetylpyridinium chloride to confirm that the bone marrow mesenchymal stem cells in the autophagy occurs,osteogenic ability can be improved.Conclusion In the presence of titanium particles,resveratrol may enhance the osteogenic differentiation of bone marrow mesenchymal stem cells by activating autophagy. |
