Effects Of Dexmedetomidine On Humoral Immunity Function In Sepsis Mice

Objective:To investigate the effects of dexmedetomidine on humoral immunity function in sepsis mice.Methods:Ninety SPF healthy male BALB/c mice,one month old,weighting 18-22 g,were randomly divided into 3 groups（n=30）:Sham operation group（group S）,sepsis group（group SEP）and dexmedetomidine group（group DEX）.Sepsis was induced by cecal ligation and puncture in groups SEP and DEX,group S under-went laparotomy.At the end of abdominal closure,the mice in DEX were injected with dexmedetomidine 30μg/kg intraperitoneally,equal volume of normal saline was given in group S and SEP.The orbital venous blood samples were collected at6,12 and 24 h(T13)after operation,and the serum concentration of IgG,IgA,IgM and complement C3 and complement C4 were detected via Elisa.After blood collecting,the spleen tissue was dissected and the proliferation of B cells was detected by MTT.Results:Compared with group S,the serum concentration of IgG、IgM and the proliferation of spleen B cells were decreased at T2,3,the proliferation of B cells of spleen was increased at T1,the serum consentration of complement C3 and complement C4 were increased at T133 in group SEP and group DEX（P<0.05）,while there was no significant changes in serum IgA concentration（P>0.05）;Compared with group SEP,the serum concentration of IgG,IgM were increased at T2,3,the proliferation of B cells of spleen was increased at T1,2,the serum consentration of complement C3 and complement C4 were decreased at T133 in group DEX（P<0.05）,there was no significant changes in serum IgA concentration（P>0.05）.Conclusion:Dexmedetomidine can improve humoral immune function in sepsis mice.
