| Objective: Polycystic ovarian syndrome(PCOS)is the most common reproductive endocrine disease in women.Overweight/obesity can aggravate the metabolic disorders and reproductive disorders of PCOS women.Obesity and visceral fat accumulation are associated with chronic hypo-inflammatory status,PCOS patients are in a chronic inflammatory state,inflammation can lead to insulin resistance(IR),and IR will be lead to hyperandrogenism(HA)and increased androgen.IR and inflammation interact with each other,becoming a vicious cycle.Traditionally,adiposities are divided into white adiposities and brown adiposities.White adipose tissue(WAT),which is mainly distributed in the subcutaneous and viscera of the body,and strongly correlated with inflammation,IR and type 2 diabetes mellitus(T2DM).Brown adipose tissue(BAT)can increase energy consumption and generate heat,mainly in the scapular,surrounding and aortic deposits.BAT can improve the energy consumption of PCOS and glucose metabolism,reduce the level of related hormones,help to improve the menstrual cycle of PCOS patients and ovulation.To investigate whether berberine(BBR)can affect polycystic ovarian syndrome(PCOS)by promoting the white fat browning.Methods: 40 female C57 BL / 6J mice,3 weeks old,were divided into blank control group,model group,low dose BBR group,middle dose BBR group and high dose BBR group,and subcutaneous injection of dehydroepiandrosterone(DHEA)for 20 days,then intragastric administration of BBR for 14 days.Observing body weight,fat wet weight and ovarian morphological changes.Detecting the levels of testosterone(T),luteinizing hormone(LH),tumor necrosis factor α(TNF-α),interleukin 6(IL-6)、bone morphogenetic protein-7(BMP-7),and uncoupling protein 1(UCP1)m RNA in adipose tissue And UCP1 protein relative expression level.Results: BBR significantly reduced the body weight,total fat,subcutaneous adipose tissue(SAT)and visceral adipose tissue(VAT)in PCOS mice.The relative expression of BMP-7、UCP1 m RNA and UCP1 protein levels in adipose tissue were significantly increased;serum T,LH levels were decreased;the number of cystic dilated follicles in mice decreased,luteal appear,ovulation disorders gradually restored.The ability of BBR to promote fat conversion to improve PCOS-related symptoms was positively correlated with the dose administered.Conclusion: BBR promoted the browning of white fats and activated endogenous BAT,resulting in decreased body weight and reduced total fat,SAT,and VAT in mice,and improved PCOS model mice HA,ovulation disorders and inflammatory conditions. |
