Ginkgo biloba extract has been widely used in the fields of medicine,health protection,skin protection and food.With the improvement of market demand and the increasing demand,improving the purity and water solubility of the active components in Ginkgo biloba extract has become the focus of scientific research.However,the methods used to improve the purity and water solubility of the active components in Ginkgo biloba extract in the past had not been suitable for industrial development.In order to develop a method which is easy for industrialization,the anti-solvent recrystallization method was applied to improve the purity and water solubility of the active components in Ginkgo biloba extract,and the experimental results were studied in detailIn this paper,the content of active components in Ginkgo biloba extract was successfully improved by the anti-solvent recrystallization method.The optimum conditions of the purification process were optimized by single factor experiment,and the optimum conditions are as following: the drug concentration was 45 mg/m L,the volume ratio of anti-solvent to solvent was 20,the stirring time was 1 min and the reaction temperature was 25 ℃.The content of ginkgetin and bilobalide were improved from 23.60% and 6.23% in the original drug to 53.85% and 11.40% in the purified powder,respectively,and the content of ginkgolic acid in the purified powder was less than 10 ppm,which meets the safety standards.In addition,the water soluble nano-powder of Ginkgo biloba extract was also prepared by the anti-solvent recrystallization method in this paper.The optimal preparation conditions conformed through the single factor method are as following: the drug concentration was 5 mg/m L,the volume ratio of anti-solvent and solvent was 20,the concentration of surfactant was 5 mg/mL,the reaction temperature was 20 ℃,the droplet acceleration was 7 mL/min,the stirring time was 1 min and the stirring speed was 900 rpm.Under the optimum conditions,the average particle size of the prepared nano-powder was 75.2 nm.The results of SEM,FTIR,XRD,DSC and TG detection show that the chemical structure and crystal form of Ginkgo biloba extract and nano-powder have no change.The dissolution rate of the nano-powder in gastric juice was significantly improved compared with that of the original drug,and the percentage for dissolution of flavonoids was increased from 63.9% to 83.49%,the percentage for dissolution of terpene lactones was increased from 23.68% to 39.32%.Moreover,the bioavailability of the nano-powder in rats was significantly higher than that of the original drug,and the bioavailability AUC value of flavonoids and flavonoids in nano-powder was 1.96 times and 1.72 times higher than that of original drug in rats respectively.
