Gel patch is a new formulation researched and developed in recent years.Compared with traditional preparations,it has the characteristics of large drug loading capacity,good penetration and sustained release effect,strong skin affinity,small toxic side effects,use conveniently,etc.Burns healing is still a huge challenge in modern medicine.Natural Plant Antimicrobial Solution(PAMs)is a traditional Chinese medicine compound product originating from Yunnan Pu’er Danzhou Pharmaceutical Co.Ltd.It has many functions such as antibacterial,anti-inflammatory,analgesic,blood stasis,promoting wound healing,etc.The product is inconvenient to carry and transport,also has a short application time on the affected area and cannot be fully utilized.PAMs was prepared into Hydrogel patches,and the mechanism on scald remediation in rats was preliminarily explored.It is not only convenient to carry and use,but also can effectively prolong the action time and has good repair capacity on scald.Firstly,in this paper,investigated the different effects of preparation conditions,matrix composition and drug addition sequence on the hydrogel performance through single-factor experiments,the results show that the best preparation conditions is 24 hours at 50 °C and 2days at room temperature.The concentration of gel skeleton Viscomate NP-700 was positively correlated with the drug loading.The concentration of the crosslinker aluminium glycinate,pH regulator tartaric acid were positively correlated with the gel strength.The concentration of cross-linking regulator EDTA was negatively correlated with the gel strength.The concentration and order of addition of drug did not affect the gel formation.On this basis,orthogonal experiments were conducted.Using NP-700,tartaric acid,EDTA,and transdermal enhancers(azone,menthol,and propylene glycol)as influencing factors,examined 3 concentrations.The hydrogel appearance,rheological properties,transdermal drug release performance in vitro and repair effect of scald in rats as the detection index.It was found that the influence degree of each factor under the selected concentration level was: tartaric acid> EDTA> NP-700> transdermal enhancer.The optimal matrix formulation for the patch is glycerol: NP-700: aluminum glycerol: PAMs: water: tartaric acid: azone: propylene glycol: SDS=30:4:0.4:31.7:27.2:0.2:3:3:0.5.And then,using the optimized matrix formulation,the efficacy and mechanism on scald remediation were studied by rat scald model.Compared the HE pathological section and the serum EGF levels by ELISA method in scalded rats between 7th and 16 th doses of rats.It was found that this hydrogel patch had good repair capacity on scald,and there were a lot of collagen fibers and capillary proliferation under the skin.In the early stage of wound healing,EGF was higher than the model group to promote wound healing.In the middle and later stages of wound repair,the down-regulation of EGF expression to decreased the scar formation.The organ index of rat indicates that there is almost no toxic or side effects.In summary,a PAMs hydrogel patch was successfully prepared,and the performance of the gel was comprehensively evaluated based on its appearance,rheological properties,cumulative release and repair capacity on scald.Tish patch has good moldability and repair capacity on scalded rats.This method both in vitro and in vivo evaluation provides an idea for the selection and optimization of patch matrix prescriptions.It also has important reference value for establishing hydrogel patch specifications,feasible measurement indicators and evaluation systems. |
