Long Noncoding RNA MINCR Regulates Cellular Proliferation,Migration,and Invasion In Hepatocellular Carcinoma

Objective:More and more experimental evidences suggested that long non-coding RNAs(lncRNAs)are aberrantly expressed in numerous cancers,including hepatocellular carcinoma(HCC).Up to date,according to a previous study,lncRNA MYC-induced long non-coding RNA(MINCR)is highly expressed in gallbladder cancer and Burkitt’s lymphoma.This report explores the effect of long non-coding MINCR on the proliferation,migration and invasion of hepatocellular carcinoma.To detect the effect of MINCR on the proliferation,migration and invasion of human hepatocellular carcinoma cells by down-regulate of MINCR by siRNA interference and to explore whether MINCR is involved in the proliferation,migration and invasion of hepatocellular carcinoma cells.It may also provide new research directions for the early diagnosis and treatment of hepatocellular carcinoma.Methods:The experimental specimens were obtained from the Second Affiliated Hospital of Nanchang University.After surgery,the patients were diagnosed as hepatocellular carcinoma by multiple pathologists(without any preoperative treatment),Tissue samples total RNA were extracted by OMEGA kit extraction.1、Quantitative real-time PCR(QPCR)was used to detect the expression of MINCR in tissues and cell lines,and the expression of MINCR was analyzed statistically.2、SiRNA downregulate of MINCR on the proliferation,migration and invasion of HepG2 and SMMC-7721 hepatocellular carcinoma cells was constructed.Cell-counting kit 8(CCK8)assay was used to detect cell proliferation ability.Scratch assay was used to detect cell migration ability.Transwell assay was used to detect cell migration and invasion ability.Results:1、The expression level of LncRNA MINCR in human hepatocellular carcinoma tissues samples was significantly higher than the corresponding paracancerous tissue samples(P <0.01).2、The expression of LncRNA MINCR in hepatocellular carcinoma cell lines was higher than that in normal liver cell line HL7702(P <0.01).3、SiRNA down regulate of MINCR expression in the HepG2 and SMMC-7721 cells were significantly reduced the proliferation,migration and invasion ability of the two hepatocellular carcinoma cells.Conclusion:1、In vitro experiments showed that downregulate of MINCR could effectively inhibit the proliferation,migration and invasion of HepG2 and SMMC-7721 cells by siRNA.2、The expression of LncRNA MINCR was positively correlated with the proliferation,migration and invasion ability of hepatocellular carcinoma cells,It may also provide new research directions for the early diagnosis and treatment of hepatocellular carcinoma.
