Emodin Inhibit Triple Negative Breast Cancer Epithelial-mesenchymal Transition And Cell Metastasis Via Wnt/β-catenin Signaling In Vitro

The migration of malignant tumor has been one of the key factors in the therapy of cancer and patients’ life quality.Especially,triple negative breast cancer is famous of migration.Emodin is one of the natural anthraquinone derivatives.And emodin has been proved that it has superior antitumor activity.This study aims to combine emodin with triple negative breast cancer and attempts to discuss whether emodin could inhibit the migration of triple negative breast cancer.At the same time,we also study the relationship between Wnt/β-catenin signal pathway and the migration of triple negative breast cancer and the role of epithelial-mesenchymal transition.This study takes the triple negative breast cancer cells(MDA-MB-468,BT549,MDA-MB-231)as the research object.First of all,triple negative breast cancer cells are incubated in medium with emodin and Wnt-C59.Then,we take the MTT assay,wound healing test,Transwell migration and invasion assay and Western Blot.And we observe the changes of cellular morphology by high-power microscope.Results indicate that emodin inhibits proliferation of triple negative breast cancer cells MDA-MB-468,BT549 and MDA-MB-231 in a does-dependence and time-dependence;Also,emodin inhibits ability of migration and invasion of triple negative breast cancer cells MDA-MB-468,BT549 and MDA-MB-231 in a does-dependence and time-dependence significantly.In addition,we observe the changes of morphology of triple negative breast cancer cells and the changes of related protein expression of EMT and the changes of related protein expression of Wnt/β-catenin signaling.The results reveals that emodin inhibits the migration and epithelial-mesenchymal transition of triple negative breast cancer via Wnt/β-catenin signal pathway.Lastly,we use Wnt-C59 to confirm that emodin inhibits the migration of triple negative breast cancer via Wnt/β-catenin signal pathway.All these results indicate that emodin inhibits the migration and epithelial-mesenchymal transition of triple negative breast cancer via Wnt/β-catenin signal pathway significantly.This study provides some basis for the anti-tumor activity of emodin,especially the inhibition of migration of breast cancer.
