Adenoid cystic carcinoma is one of the most common malignant tumor in saliary gland and often occurs in minor saliary gland.The tumor is characterized by hidden onset,slow growth,strong invasion,high rate of recurrence,distant metastasis,and it is invaid to conventional chemotherapy and radiotherapy.growing along the nerve and invading the surrounding blood vessels brings many difficulties to SACC entirely surgical resection.Since lacking of objective means to exactly locate invaded parts in surgery plus by blindly expanding resection will inevitably give patients unnecessary deformity and dysfunction,the sole surgical effect and the long-term outcome is not ideal.With the rise of gene therapy,further study of the biological characteristics of SACC,pathogenesis study in genes and protein levels,looking for the biological markers of occurrence,development and prognosis become the domestic and foreign research hotspot.Undoubtedly gene therapy will be the most potential and effective treatment for SACC in the future.The ubiquitin proteasome system(UPS)regulates degree of proteins to maintain the dynamic balance of proteins,which have a close relationship with signal transduction,cell cycle,turnover and growth of tumor.As a molecular target for new antitumor drugs,UPS have become a hot research topic.The purpose of this study was to elucidate the expression and relationship of UBE3A,USP7 and p53 in adenoid cystic carcinoma of salivary glands.The correlation of expression of UBE3A,USP7 and p53 protein in adenoid cystic carcinoma of salivary glands were also discussed to provide data support for clarifying the formation mechanism of salivary adenoid cystic carcinoma.Methods:1.In this study,we collected 20 cases of SACC fresh specimens and 20 cases of normal saliary gland tissues specimens,through the immunohistochemistry,RT-PCR and Western blot to detect the expression of UBE3A,USP7 and p53 in the gene and protein level.And analyze whether their expression level is consistent with the demographic data of SACC patients.2.RT-PCR and Western blot were used to detect the gene and protein levels of UBE3 A,USP7 and P53 in SACC cell lines.Results:1.Ubiquitin ligase UBE3A and deubiquitinating enzyme USP7 were expressed in SACC tissues,and the mRNA and protein levels are higher than that of the control group(P<0.05),indicating that UPS is active in SACC cancer tissues;UBE3A,USP7 and p53 expression correlation,suggesting that UPS may produce corresponding effects through P53 in SACC.2.Multivariate logistic regression analysis confirmed the expression of UBE3Aand USP7 expression was an independent risk factor for distant metastasis in SACC patients(P<0.05).3.Similar to the results of histological study,RT-PCR PCR and Western blot detected that both USP7 and UBE3 A were expressed in ACC-M(high lung metastatic cell line)and ACC-2(low lung metastatic cell line),but There was no significant difference in the measured values between the two cell lines(P>0.05).Conclusion:1.UBE3A,USP7,p53 were all expressed in SACC tissues and cell lines.2.UPS was active in cancer tissues.3.UBE3A and USP7 may regulate the expression of p53,the key tumor suppressor gene in SACC,in tumor cells.In order to achieve the purpose of anti-promoting/promoting tumor growth.4.There was no difference in the expression of UPS in ACC high/low metastatic cell lines,and subsequent experiments were needed to verify the correlation between UPS and the adhesion and metastasis of these tumors.The above results of study can provide new targets and directions for clinical diagnosis and treatment of SACC and have guiding significance in the selection of clinical chemotherapy drugs.
