Objective: To detect the genotypes of C677 T and A1298 C in methylenetetrahydrofolate reductase(MTHFR)gene in children with acute lymphoblastic leukemia(ALL),explore the relationship beween the two gene locus polymorphisms and the adverse reactions which treated high-dose methotrexate(HD-MTX)chemotherapy in children with ALL.Methods: Collect the peripheral blood samples of 33 our hospital’s children with acute lymphoblastic leukemia who completed remission of VDLP chemotherapy from 2015 to 2017.Statistic the genotype distribution characteristics of MTHFR C677 T and A1298 C,observed and collected the clinical data about the adverse reactions of children with ALL treated with high-dose methotrexate;and analyze for the association of each genotype with adverse events.Results: 1.MTHFR C677 T genotype distribution is mostly heterozygous CT,MTHFR A1298 C genotype distribution is mostly homozygous AA;2.The adverse reactions which be caused by HD-MTX chemotherapy focus on the bone marrow suppression and the skin mucosal damage,Followed by gastrointestinal reactions,abnormal liver function,no abnormal renal function was found in this study;3.There is no significant difference between MTHFR C677 T genotypes and bone marrow suppression、 skin mucous membrane damage、 gastrointestinal tract and abnormal liver function;But there is a significant difference in the gastrointestinal reactions between MTHFR A1298 C genotypes(P<0.05),and the risk of gastrointestinal reactions is lower in patients carrying the MTHFR 1298 AA genotype than in AC+CC.Conclusion: There is no significant relationship between the MTHFR C677 T polymorphisms and the adverse reaction of high-dose MTX chemotherapy in children with ALL;and MTHFR A1298 C polymorphism is closely related to the adverse reactions of MTX,There are significant differences in the gastrointestinal reactions.
