| Background “Airflow limitation,not completely reversible” is a common pathophysiological feature of Chronic obstructive pulmonary disease(COPD).Airway and pulmonary vascular remodeling is the key factor leading to the incomplete and reversible airflow limitation of COPD.Vascular endothelial growth factor(VEGF)and transforming growth factor-beta 1(TGF-β1)are closely related to the airway and pulmonary vascular remodeling of COPD.Resveratrol is a naturally existing polyphenol phytoalexin,it has extensive pharmacological effects,such as radical scavenging,antioxidation,anti-inflammation.Studies have shown that natural resveratrol has anti-inflammatory effects in the treatment of COPD,but there are few studies on its effect on fibrosis.Objective To investigate the expression of VEGF and TGF-β 1 in COPD rats resveratrol intervention group COPD rats and normal rats expression in lung tissue,clear the role of resveratrol respiratory function of COPD rats,and to explore the effect of resveratrol on the expression of VEGF and TGF-β1 in COPD rats,and provide laboratory basis for clinical treatment of reversing COPD pathology.Methods Thirty healthy conventionalSprague-Dawley(SD)were randomly divided into three groups: COPD group,resveratrol group,and normal group.In COPD group,the COPD rat model was established by passive smoking and intratracheal Lipopolysaccharide(LPS)instillation.In resveratrol group,rats were treated with15mg/kg.dresveratrol by gavage,1h later the rats were treated with COPD group process.In control group,rats didn’t any treatment.All three groups of rats were operated as follows:1.Observation of general condition and lung gross view.2.Observation of ultrastructure of lung tissue cells by scanning electron microscope.3.Detection of VEGF and TGF-β1 in lung tissue by enzyme-linked immunosorbent assay.4.Detection of VEGF and TGF-β1 levels in lung tissue by RT-PCRResults 1.General condition: rats in group COPD had snivela(12th day),at day 21,rats had a significant increase of snivela,and breathing amplitude increased in calmness,what’s more,rats had gasping.In resveratrol group,rats in COPD group had snivela(15th day),at day 21,rats had a significant increase of snivela,and breathing amplitude increased in motion state,only individual rats rats had gasping.In normal group,rats didn’t appear snivela,respiratory secretions,and wheezing.General view: the lungs of COPD groupwere black gray,border became blunt,markings decreased,elasticity was poor.In resveratrol group,the lungs of COPD groupwere gray white,border became more blunt,markings significantly decreased,elasticity was worse.In normal group,lungs were pink,markings were clear,elasticity was good.2.hypermicro-instructure of lung tissue: in COPD group,microvillus of type Ⅱpneumocyte decreased,empty lamellar bodys,swollen mitochondrions.In resveratrol group,microvillus of type Ⅱ pneumocyte decreased,empty lamellar bodys,but mitochondrions were normal.In normal group,the ultrastructure of lung hadnoabnormalchange.3.The results of ELISA: the protein content of VEGF in lung homogenate of resveratrol group was lower than that of COPD group(t=11.162,p=0.000),higher than that of normal group(t=9.527,p=0.023),VEGF in lung tissue homogenate of COPD group was higher than that of normal group(t=14.36,p=0.000).The protein content of TGF-β1 in lung homogenate of resveratrol group was lower than that of COPD group(t=9.281,p=0.000),higher than that of normal group(t=10.135,p=0.000),TGF-β1 in lung tissue homogenate of COPD group was higher than that of normal group(t=10.311,p=0.019).4.The results of RT-PCR: The level of VEGFmRNA in the lung homogenate of theresveratrol group was lower than that of the COPD group(t=7.029,p=0.019),higher than that of the normal group(t=9.733,p=0.003),and the lung tissue homogenate of the COPD group was higher than that of the normal group(t=7.216,p=0.000).The results of RT-PCR:The level of TGF-β1mRNA in the lung homogenate of the resveratrol group was lower than that of the COPD group(t=10.259,p=0.009),higher than that of the normal group(t=8.372,p=0.000),and the lung tissue homogenate of the COPD group was higher than that of the normal group(t=17.122,p=0.000).Conclusion 1.Resveratrol can effectively improve respiratory function of COPD rats.2.Resveratrol improves respiratory function in COPD rats by reducing VEGF and TGF-β1 expression. |
