Objective:By detecting the expression of the disintegrin metalloproteinase ADAM-like Decysin-1(ADAMDEC1)in meningioma tissue,To investigate the correlation between the expression of ADAMDEC1and clinical features in meningiomas.Method:(1).Clinical data of 50 patients whose underwent surgical resection and pathological diagnosis of meningiomas were collected from Department of Neurosurgery,Second Affiliated Hospital of Southwest Medical University from July 2015 to December 2017.All 50 patients selected that had no other neoplastic disease;They do not have cardiovascular disease,diabetes,infectious diseases and other complications;All patients underwent craniotomy for the first time,and no treatment such as radiotherapy,chemotherapy,gene therapy and immunotherapy were performed prior to surgery.Mesenteric postoperative pathological results in accordance with the WHO classification criteria for meningioma grading,including WHO grade I meningioma in 28 cases,WHO grade II meningioma in 18 cases,WHO grade III meningioma in 4 cases;According to the classification of invasive meningioma diagnostic criteria,34cases of invasive meningioma,noninvasive meningioma in 16 cases;According to the dissection of meningioma at the base of the attachment site,divided into general parts of meningioma(convex meningioma and cerebellar meningioma)in 24 cases and skull base meningioma(skull base central meningioma and skull base non-central meningioma)26 cases;According to the preoperative imaging data and postoperative pathological results,there were 11 cases of calcified meningiomas and 39 cases of non-calcified meningiomas.(2).Twelve patients with acute severe traumatic brain injury were collected from July 2015 to December 2017.(3).Emergency line of intracranial decompression surgery was collected normal brain tissue samples.Using immunohistochemistry(S-P)method,Detection of ADAMDEC1 protein in different types of meningioma and normal brain tissue expression levels,The relationship between the positive expression and clinical features of ADAMDEC1 in meningioma was statistically analyzed.Results:(1)The positive expression of ADAMDEC1 in meningioma tissues mainly concentrated in the cell membrane and cytoplasm.The normal brain tissue showed no significant positive expression.ADAMDEC1 protein expression in meningioma and normal brain tissue:38 cases of ADAMDEC1 protein were positive in 50 cases of meningiomas,12 cases were negative expression,In 12cases of normal meningeal tissue,11 cases of ADAMDEC1 protein expression was negative,1 case was positive.The expression of ADAMDEC1 protein in meningioma tissues and the expression of ADAMDEC1 protein in normal meningeal tissues were statistically analyzed,The difference was statistically significant(Z=-4.000,P<0.05).(2)The expression of ADAMDEC1 protein in WHO I WHO II and WHO III meningiomas:10 of the 28 WHO grade I meningiomas were negative(-),8 were weakly positive(+),6 were positive(++)and 4 were strong positive(+++).In 18 cases of WHO grade II meningioma tissues,2 were negative(-),5 were weakly positive(+)and 5 were positive(++),6 were strong positive(+++).In 4 cases of WHO grade III meningiomas,0 was negative(-),0 was weakly positive(+)and 2 was positive(++),2 cases showed strong positive(+++).The expression of ADAMDEC1 in WHO gradeⅠmeningioma and the expression of ADAMDEC1 in normal meningioma were statistically analyzed,The difference was statistically significant(Z=-3.188,P<0.05);The expression of ADAMDEC1 in WHO grade II meningiomas was compared with the expression of ADAMDEC1 in normal meningeal tissues,the difference was statistically significant(Z=-4.130,P<0.05).The expression of ADAMDEC1 in WHO grade III meningiomas was compared with the expression of ADAMDEC1 in normal meningeal tissues,the difference was statistically significant(Z=-3.546,P<0.05).The expression of ADAMDEC1 in WHO gradeⅠmeningioma and the expression of ADAMDEC1 in WHO gradeⅡmeningioma were statistically analyzed,the difference was statistically significant(Z=-2.070,P<0.05);The expression of ADAMDEC1 in WHO gradeⅠmeningioma and the expression of ADAMDEC1 in WHO grade III meningioma were statistically analyzed,the difference was statistically significant(Z=-2.242,P<0.05);The expression of ADAMDEC1 in WHO grade II meningioma and the expression of ADAMDEC1 in WHO grade III meningioma were statistically analyzed,The difference was no statistically significant(Z=-1.161,P>0.05).The WHO grade of meningioma and ADAMDEC1 expression intensity correlation analysis,the two was positively correlated(r_s=0.390,P<0.05).(3)The expression of ADAMDEC1proteinininvasivemeningiomaandnoninvasive meningioma:There were 34 cases of invasive meningioma and 16 cases of non-invasive meningiomas in 50 cases of meningioma,The expression of ADAMDEC1 protein in the invasive meningioma group and the expression of ADAMDEC1 protein in the non-invasive meningiomas were statistically analyzed,the difference was statistically significant(Z=-4.919,P<0.05);In the28 cases of WHO gradeⅠmeningioma,there were 12 cases of invasive meningioma,16 cases of noninvasive meningioma,The expression of ADAMDEC1 in invasive meningioma of WHO grade I meningiomas and in non-invasive meningiomas were statistically analyzed,the difference was statistically significant(Z=-4.357,P<0.05).The correlation between the expression of ADAMDEC1 protein and the invasiveness of meningiomas was positively correlated(r_s=0.618,P<0.05).(4)The expression of ADAMDEC1protein in the skull base central area and skull base non-central meningioma tissue:There were 26 cases of skull base meningioma in 50 cases of meningiomas,Including the central skull base meningioma in 12 cases,14 cases of non-central skull base meningioma,The expression of ADAMDEC1 in the central skull base meningiomas and the expression of ADAMDEC1 in the non-central skull base meningiomas were statistically analyzed,The difference was no statistically significant(Z=-0.081,P>0.05);Among 24 cases of meningioma in general,11 cases of cerebellar meningioma,13 cases of convexmeningioma,ADAMDEC1 expression in brain meristerema,and ADAMDEC1 expression in meningioma were statistically analyzed,The differencewasnostatisticallysignificant(Z=-0.097,P>0.05).(5)The expression of ADAMDEC1 protein in calcified meningiomaand non-calcified meningioma:In 50 cases of meningiomas,CT findings of 11 cases and postoperative pathological findings suggest calcification,39 cases without calcification,ADAMDEC1 expression in calcified meningioma tissue and ADAMDEC1 expression in non-calcified meningioma tissue were statistically analyzed,The difference was no statistically significant(Z=-1.391,P>0.05).(6)Expression of ADAMDEC1 Protein in Meningioma of different Sex and female patients of different Ages:Among the 50 patients with meningioma,there were 18 males and 32 females,the ADAMDEC1 expression in meningioma of male patients and ADAMDEC1 expression in meningiomas of female patients were statistically analyzed,The difference was no statistically significant(Z=-0.783,P>0.05).Among 32 women with meningiomas,18 cases of meningioma patients aged 50 or older,There are 14 women with meningiomas less than 50 years old,The expression of ADAMDEC1 in meningioma of women aged 50 or older and the expression of ADAMDEC1 in meningioma of women aged less than 50 years oldwere statistically analyzed,The difference was no statistically significant(Z=-1.164,P>0.05).Conclusion:⑴ADAMDEC1 protein expression in meningioma tissue was significantly higher than normal meningeal tissue,ADAMDEC1 protein may be involved in the formation ofmeningioma.⑵ADAMDEC1 protein was expressed in different grades(WHO grade)meningiomas,and the higher the WHO grade level,the stronger its expression intensity,indicating that ADAMDEC1 protein may be associated with the WHO classification of meningioma.⑶ADAMDEC1 protein expression in invasive meningioma tissue was significantly higher than non-invasive meningioma tissue expression intensity;The expression of ADAMDEC1 protein in invasive meningioma in WHO gradeⅠmeningioma group was significantly higher than that in non-invasive meningioma group in WHO gradeⅠmeningioma,indicating that ADAMDEC1 protein may have a positive correlation with the invasiveness of meningioma.⑷There was no significant difference in the expression of ADAMDEC1 protein in meningiomas of men and women with different ages and gender,indicating that ADAMDEC1protein may not be associated with meningioma in different age groups and female patients.⑸ADAMDEC1 protein in the skull base central meningioma and skull base non-central meningioma,convex meningioma and cerebral parenchyma meningioma tissue expression no significant difference;The expression of ADAMDEC1 protein in the calcified meningioma and non-calcified meningioma had no significant difference,indicating that the ADAMDEC1 protein might be related to the different parts of the tumor and meningiomas There is no correlation between.
