Objective:To investigate the expression level of BATF and IRF-4 in decidua in normal pregnant women and patients with recurrent spontaneous abortion.Compare and anaylse the difference between the two groups.Methods:Samples of normal pregnant women and patients with recurrent spontaneous abortion in Renmin Hospital of Wuhan University was collected from 2015 to 2017.Some Samples were frozen and stored,others were fixed and then embedded in paraffin.The frozen samples were performed with quantitative Real Time-Polymerase Chain Reaction to detect the mRNA expression level and Western Blot to detect the protein expression level of BATF and IRF-4 respectively.The praffin-embedded samples were performed with Immunohistochemistry to detect the location of BATF and IRF-4 protein in decidua.The expression level of BATF and IRF-4 were observed and the difference between the two groups was compared and analysed.Results:1.The expression level of BATF and IRF-4 could be observed in decidua in normal pregnant women and patients with recurrent spontaneous abortion.2.Compared with normal pregnant women,the mRNA(1.00±0.10vs 3.18±0.27)and protein level(0.03±0.01 vs 0.622±0.03)of BATF were increased significantly in patients with recurrent spontaneous abortion(P<0.05).3.BATF protein could be observed both in nucleus and cytoplasm decidua.4.Compared with normal pregnant women,the mRNA(1.00±0.10 vs 2.91±0.52)and protein level(1.16±0.01 vs 1.49±0.03)of IRF-4 were increased significantly in patients with recurrent spontaneous abortion(P<0.05).5.Compared with normal pregnant women,the rate of BATF/IRF-4 protein level(0.03±0.001 vs 0.42±0.02)was increased significantly in patients with recurrent spontaneous abortion(P<0.05).6.IRF-4 protein was present in the nucleus mainly.Conclusion:1.This study first discovered the expression of BATF and IRF-4 in decidua in normal pregnant women and patients with recurrent spontaneous abortion.2.The expression level of BATF and IRF-4 in patients with recurrent spontaneous abortion are increased abnormally,which may contribute to the development of recurrent spontaneous abortion.This finding provides a new aspect in etiology which may contribute to unexplained recurrent spontaneous abortion.3.Nuclear translocation of BATF might occur in decidua.
