ObjectiveProducers of high and low metastatic potential for hepatocellular carcinoma in nude mice animal models,to observe the effect of Bone marrow mesenchymal stem cells(BMSCs)on tumor growth in animal models of hepatocellular carcinoma with high and low metastatic potential,and to detect the expression of metastasis-related factors and apoptosis-related factors in animal model specimens.MethodsProducer animal model of high metastatic potential hepatocellular carcinoma(MHCC97-H)and animal model of low metastatic potential hepatocellular carcinoma(MHCC97-L),the animals of MHCC97-H and MHCC97-L groups were randomly divided into two groups.Namely MHCC97-H model experimental group,control group,MHCC97-L model experimental group and control group.On the 7th day after tumor inoculation,in the experimental group,BMSCs were injected into the caudal vein with a dose of 5×105/mice,the control group was injected with BMSCs medium 0.2ml/mice.The subcutaneous tumor volume was measured with a caliper every 3 days after the start of the experiment,the animals were sacrificed on the 13 th day,16 th day,19 th day,22 th day,25 th day,28 th day,31 th day,34 th day,37 th day and 40 th day after tumor inoculation,the tumor tissues were completely taken and weighed,calculating the tumor inhibition rate.Producer animal model of low metastatic potential hepatocellular carcinoma,the MHCC97-L animal were randomly divided into experimental group and control group,the experimental group and the control group were treated with the same way as above.The difference is,the volume of the subcutaneous tumor was measured once a week after the start of the experiment,the animals were sacrificed at the 2nd,3rd,4th,5th and 6th week after tumor inoculation,the tumor tissues were completely taken and weighed,calculating the tumor inhibition rate.Real-time PCR was used to detect the expression of metastasis-related factors,including osteopontin,Bone sialoprotein,IntegrinαV gene and the expression of apoptosis-related factors including Bcl-2,Bax and caspase3 gene.ResultsBMSCs showed no inhibitory effect on tumor weight and volume in MHCC97-H animal model,the tumor weight and volume in the experimental group were always higher than those in the control group.statistical analysis showed that the tumor weight and volume difference between the experimental group and the control group were statistically significant at the 13 th day;BMSCs showed an inhibitory effect on the weight and volume of MHCC97-L tumor,statistical analysis showed that the tumor volume difference between the experimental group and the control group were statistically significant at the 22 th day,the experimental group was higher than the control group,the rest of the time showed that the experimental group of tumor weight and volume were lower than the control group.BMSCs showed an inhibitory effect on the weight and volume of MHCC97-L,the tumor weight and tumor volume in the experimental group were always lower than those in the control group,statistical analysis showed that the tumor weight difference between the experimental group and the control group was statistically significant at the third week,the tumor weight of the experimental group was significantly lower than that of the control group;The difference of the tumor volume between the experimental group and the control group was statistically significant at the 2nd,3rd,5th and 6th weeks.The tumor volume of the experimental group was significantly lower than that of the control group.The expression of metastasis-related factors OPN,BSP and IntegrinαV in the experimental group were significantly lower than that in the control group,the expression of pro-apoptotic related factors Bax and caspase3 were increased after BMSCs intervention,and the expression of anti-apoptotic related factor Bcl-2 was decreased gradually.ConclusionsBMSCs had no inhibitory effect on the growth of MHCC97-H and had an inhibitory effect on the growth of MHCC97-L.The inhibitory effect of BMSCs on tumor weight and volume was the most obvious at the 3rd week,and the inhibitory effect was gradually weakened with the passage of time.
