| ObjectiveThe aim of this study was to investigate the expression of Toll-like receptor 4(TLR4),myeloid differentiation factor 88(MyD88)and nuclear factor kappa B(NF-κB)in colorectal cancer and the correlation between the three for the diagnosis and treatment of colorectal cancer laid the theoretical foundation.MethodsThe expressions of TLR4,MyD88 and NF-κB in 70 cases of colorectal cancer,36 cases of colorectal adenoma and 20 cases of adjacent normal mucosa were detected by immunohistochemistry PV6000.The expression of TLR4,MyD88 and NF-κB were analyzed by clinical method relationship of pathological indicators.The expression of TLR4,MyD88 and NF-κB in 30 cases of fresh colorectal cancer tissues and corresponding 5cm intestinal mucosa were detected by western blot.The correlation and clinical significance were analyzed.ResultsThe expressions of TLR4,MyD88 and NF-κB in colorectal carcinoma were significantly higher than those in colorectal adenocarcinoma and adjacent normal mucosa(P<0.05)by immunohistochemistry.The expressions of TLR4,MyD88 and NF-κB There were significant correlation with tumor differentiation,lymph node metastasis and TNM stage(P<0.05),but not with the age,sex and tumor location(P>0.05);the expression of TLR4 was correlated with theexpression of MyD88 and NF-κB(P<0.05).The expression of TLR4,MyD88 and NF-κB in 30 cases of colorectal cancer was higher than that in the adjacent 5cm intestinal mucosa(P<0.05).ConclusionsThe overexpression of TLR4,MyD88 and NF-κB in colorectal cancer correlated with tumor cell differentiation,lymph node metastasis and TNM staging,suggesting that TLR4 was involved in the differentiation,invasion and metastasis of tumor cells.TLR4 expression was correlated with MyD88 and NF-κB analysis showed a positive correlation,suggesting that TLR4 overexpression can promote MyD88,NF-κB expression increased. |
