Objective:This experiment takes the natrual menopausal rats as research subject,to investigate the changes of apoptosis factors bcl-2 and Caspase-3 in ovary,treated the rats with the QXZST or Remifemin,then observe the effect of QXZST and Remifemin on bcl-2 and Caspase-3.Method: 1.The rats 13 to 15 months were randomly classified into menopause group,Remifemin group,QXZST low-dose group,moderate-dose group,high-dose group,the rats 4month as young group.2.All of the rats administered by gavage for 4 weeks,then extracted the ovaries and blood.3.To weigh the ovaries,use ELISA means measure the levels of serum E2,FSH,LH.4.To use Western blot measure the expression of bcl-2 and Caspase-3 protein.5.To use PT-PCR measure the expression of bcl-2 and Caspase-3mRNA.Results: 1.Compared with young group,the ovary weight of menopause group obvious reduce(P < 0.05);the ovary weight of the QXZST groups and Remifemin group compared with the menopause group have no statistical significance.2.Compared with young group,the E2 level drop,FSH and LH level rise of menopause group(P<0.05);the hormone levels of the QXZST groups and Remifemin group compared with the menopause group have no statistical significance.3.Compared with young group,the expression of bcl-2 protein and bcl-2 mRNA reduced of menopause group(P < 0.05);compared with the menopause group,the expression of bcl-2protein and bcl-2 mRNA increased of QXZST high-dose group and Remifemin group(P<0.05).4.Compared with young group,the expression of Caspase-3 protein and Caspase-3 mRNA increased of menopause group(P < 0.05);compared with the menopause group,the expression of Caspase-3 protein and Caspase-3 mRNA reduced of QXZST high-dose group and Remifemin group(P<0.05).Conclusion: 1.QXZST and Remifemin can fine tuning the hormone levels of menopausal rats to a new equilibrium,but not significantly.These results indicate that QXZST and Remifemin can improve the HPG disorder to some degree.2.In the mitochondrial apoptotic pathways,QXZST and Remifemin not only affect the upstream bcl-2 to avoid apoptosis but also act on downstream Caspase-3 to prevent the irreparable damage of cell.
