【Objective】: To explore the expression and effect of allograft inflammatory factor-1(AIF-1),matrix metalloproteinase 9(MMP-9),and cyclooxygenase-2(COX-2)in human ruptured intracranial aneurysm tissues.【Methods】: To collect 12 cases of ruptured intracranial saccular aneurysms as the experimental group and superficial temporal arteries as control group.The microstructure of the specimens were observed by optical microscope.Immunohistochemical staining was used to detect the expression of AIF-1,MMP-9 and COX-2 in the experimental group and the control group.AIF-1,MMP-9 and COX-2 expression were quantitatively analyzed by Image-Pro-Plus 6.0 software.Spearman was used to analyze the correlation of AIF-1、MMP-9 and COX-2 expression in the experimental group.【Results】1.The damage,disappeared or degeneration of endothelial cells(ECs)and tumor thrombosis were observed in experimental aneurysm tumor specimens.The vascular smooth muscle cells(VSMCs)of tumor wall medial membrane is degenerated or disappears.The whole of tumor wall observed inflammatory cells infiltration(lymphocytes and macrophages),mainly existing with the endometrium and medial membrane.In the control group,the microstructure of the superficial temporal arteries were intact,the middle membrane was thick and inflammatory cells infiltration was not observed.2.Immunohistochemical staining showed that the intima and middle of ruptured aneurysm tumor tissues presented a lot of positive areas,suggesting that AIF-1,MMP-9 and COX-2 mainly expressed in macrophages and lymphocytes of the aneurysm inner and middle layers,not in the superficial arteries.The expression levels of AIF-1,MMP-9 and COX-2 in ruptured intracranial aneurysm were 0.0118 ± 0.0097,0.0038 ± 0.0028 and 0.0044 ± 0.0037,respectively,and the temporal superficial expression levels of AIF-1,MMP-9 and COX-2 were 0.0000,0.0000 and 0.0000 respectively.The difference between the two groups was significant(P<0.01),indicating that the expression levels of AIF-1,MMP-9 and COX-2 in ruptured intracranial aneurysm specimens obviously high than superficial temporal arteries(P=0.014 <0.05).3.The positive correlation between AIF-1 and MMP-9 was statistically significant(R=0.685,P=0.014<0.05),while AIF-1 and COX-2,MMP-9 and COX-2 were not statistically obvious correlation.4.Analysis the relationships between AIF-1,MMP-9,COX-2 expression and sex,smoking in the experimental group.The results shown that average level of AIF-1 in male(0.016 ± 0.013)was higher than in female(0.009 ± 0.006)with statistically significant(T = 1.440,P = 0.043 <0.05),but had no significant correlation with smoking.【Conclusion】1.AIF-1 is highly expressed in the wall of ruptured intracranial saccular aneurysms,mainly expressed in macrophages and lymphocytes,might be a risk factor for IA.2.The expression of AIF-1,MMP-9 and COX-2 increased in IA tissues,they may participate in the formation and progression of IA.
