| Objective1 The purpose of the present study is to investigate the effects of the electroacupuncture at Baihui,Fengfu,Shenshu on the cognitive function and learning memory in model mice with radiation induced brain injury.2 To investigate the effect of electroacupuncture at "Baihui","Fengfu" and "Shenshu" on proliferation and differentiation of neural stem cells in hippocampus of mice with radiation induced brain injury and its protective mechanism.MethodsC57BL/6J(30-day old)mice were randomly divided into 10 groups,the control group,model groups(1week,2weeks,3weeks),electroacupuncture-nonacupiont(E-non A)groups(1 week,2weeks,3weeks)and electroacupuncture-acupiont(EA)groups(1week,2weeks,3weeks),10 mice for each group.Then the mice were irradiated(IR)on the cerebral hemisphere with 8 Gy.Take difference courses of electro-acupuncture as treatment method.At the beginning of acupuncture treatment,each mouse was injected intraperitoneally with Brd U,and the body weight was measured.By using open-field test in cognitive function,Morris water maze to test the spatial learning and memory ability.Immunohistochemical method and immunofluorescence double labeling method were used to detect the proliferation(Brd U)and differentiation(Brd U/Neu N,Brd U/GFAP)of neural stem cells in hippocampus of mice with radioactive brain injury.Results1 Weight: On the first day of modeling,the weight of mice in each group was not statistically significant(P>0.05).On day 7,compared with the control group,the weight of mice in model groups(2w,3w)were decreased significantly(P<0.05);Compared with model group(2w),the weight of the EA group(2w)was significantly increased(P<0.05);Compared with model group(3w),the weight of the EA group(3w)was significantly increased(P<0.05).On day 14,the weight of mice in each group was statistically significant(P<0.05).Compared with model group(2w),the weight of the EA group(2w)was significantly increased(P<0.05);Compared with model group(3w),the weight of the EA group(3w)was significantly increased(P<0.05).On day 21,the weight of mice the model group(3w)was statistically significant(P<0.05).Compared with model group(3w),the weight of the EA group(3w)was significantly increased(P<0.05).2 Open field test:(1)Compared with the control group,the modification times of model group(1w)were significantly decreased(P<0.01).Compared with model group(1w),the rearing times of the EA group(1w)were significantly increased(P<0.05).(2)Compared with the control group,the modification times and the rearing times of model group(2w)were significantly decreased(P<0.01).Compared with model group(2w),the rearing times,modification times and crossing times of the EA group(2w)were significantly increased(P<0.05).(3)Compared with the control group,the rearing times,crossing times and the modification times of model group(3w)were significantly decreased(P<0.05).Compared with model group(3w),the rearing times,modification times and crossing times of the EA group(3w)were significantly increased(P<0.01).3 Morris water maze test:(1)Compared with the control group,the latent period of third days of model group(1w)was significantly increased(P<0.01),and the crossing times and the time spending on target area were decreased significantly(P<0.05).Compared with model group(1w),the latent period of EA group(1w)was decreased,but there was no statistical significance.And the time spending on target area was decreased significantly(P<0.01).(2)Compared with the control group,the latentperiod of model group(2w)was significantly increased(P<0.01),and the crossing times and the time spending on target area were decreased significantly(P<0.05).Compared with model group(2w),the latent period of EA group(2w)were significantly decreased(P<0.01),and the crossing times as well as time spending on the target area were decreased significantly(P<0.05).(3)Compared with the control group,each day of the latent period of model group(3w)were significantly increased(P<0.05),and the crossing times and the time spending on target area were decreased significantly(P<0.05).Compared with model group(3w),the latent period of EA group(3w)were significantly decreased(P<0.01),and the crossing times as well as time spending on the target area were decreased significantly(P<0.05).4 Neural stem cell proliferation: There were appearanced Brd U markers in the hippocampus of mice in each group,indicating that neural stem cells proliferation.Compared with the control group,the OD of each model groups(1w,2w,3w)were significantly decreased(P<0.05).Compared with model group(1w),the OD of EA group(1w)was decreased,but has no statistical significance.Compared with model group(2w),the OD of EA group(2w)was significantly increased(P<0.05).Compared with model group(3w),the OD of EA group(3w)was significantly increased(P<0.05).5 Neural stem cell differentiation: Each groups showed Brd U/Neu N,Brd U/GFAP in the hippocampus of mice in each group,indicating that neural stem cells differentiation.Compared with the control group,the positive cells of each model groups(1w,2w,3w)were significantly decreased(P<0.05).Compared with model group(1w),the positive cells with EA group(1w)was significantly increased(P<0.05).Compared with model group(2w),the positive cells with EA group(2w)was significantly increased(P<0.01).Compared with model group(3w),the positive cells with EA group(3w)was significantly increased(P<0.05).Conclusion1 EA acupuncture "Baihui" and "Fengfu" and "Shenshu" to improve the damage of cognitive function in the radiation induced brain injury mice,and increases the effect of acupuncture acupuncture treatment is more obvious;2 EA acupuncture "Baihui" and "Fengfu" and "Shenshu" to improve the damage of learning memory in the radiation induced brain injury mice.And with the increase of EA acupuncture treatment,the ability of learning memory was further improved.3 EA acupuncture "Baihui" and "Fengfu" and "Shenshu" can promote neural stem cell proliferation and differentiation in the hippocampus of the radiation induced brain injury mice. |
