Detection Of Circulating MiRNA-4741 And MiRNA-6086 In Hepatocellular Carcinoma And Its Clinical Significance

Purpose: The purpose of this study was to detect the expression of serum miRNA-6086,miRNA-4741,miRNA-122 a in normal subjects and patients with HCC,and to study the relationship between three miRNAs in serum and the diagnosis,prognosis of patients with liver cancer.Methods: Collected from December 2014 to December 2015 in hospital of south China and First hospital affiliated to university of south China who have been diagnosed and treated 45 cases of liver cancer patients into the study group,the same period in hospital of south China physical examination of the 45 healthy people into the control group;The miR-122 a,miR-6086 and miR-4741 in serum were detected by real-time PCR method,and the results were statistically analyzed.Result: 1)MiRNA-122 a of Hepatoma group in the experimental was lower than that of the control group,but the content of miR-6086 and miRNA-4741 was higher than that of the control group and the difference both were statistically significant(P > 0.05).2)Statistical analysis revealed that there were no significant differences in the relative expression of the patients in different age,stage,classification,tumor size,AFP level,liver function(ALT,AST,TB)and so on(P > 0.05).3)ROC curve analysis:AUC of MiRNA-122 a area was greater than the AUC of miRNA-6086 and miRNA-4741,where the miRNA-4741 AUC area was smallest,but all of them were statistically significant;4)The specificity of MiR-6086 and miR-122 a combined with AFP in the diagnosis of primary liver cancer was significantly higher than before,and the sensitivity was not significantly changed,The sensitivity of miRNA-4741 combined with AFP was slightly increased and the specificity was not changed;MiRNA-4741 combined with miRNA-6086 and miRNA-4741,miRNA-6086 combined with AFP,the specificity was significantly lower than that of the single analysis,and the sensitivity increased slightly,(P > 0.05);5)The higher expression level of miRNA-6086 and miRNA-4741,HCC patients have shorter of survival time;Instead,when the expression level is lower,HCC patients have longer of survival time,(P < 0.05),Although the expression level of miRNA-122 a was down regulated,the lower expression of patients have longer survival time than the higher expression survival time,(P > 0.05).Conclusion: 1.The expression levels of serum Mi RNA-6086 and miRNA-4741 in HCC patients were significantly higher than those in normal controls,On the contrary,the expression level of miRNA-122 a was significantly lower than that of normal subjects and the three may be a potential tool for the diagnosis and prognosis of HCC.2.MiRNA-6086 and miRNA-122 a combined with AFP analysis can obviously improve the diagnostic specificity of HCC patients.3.MiRNA-6086,miRNA-4741 and miRNA-122 a were related to the duration of survival,and the higher the expression of the three,the shorter the patient’s survival time.
