Effect Of Jiedu Tongluo Baoshen Method On The Expression Of Bax,Bcl-2,Caspase-3 Of Apoptosis Related Factors In Rats With Diabetic Nephropathy

Purpose:This experiment mainly observe the effect of Jiedu Tongluo Baoshen Method to apoptosis factors Bax,Bcl-2,caspase-3 of the podocyte of experimental DM rats’kidney,study the Jiedu Tongluo Baoshen Method for protection mechanism of podocyte in Diabetic Nephropathy.Method:60 Wistar rats with half male and half females,give high sugar high-fat feed.All the rats were fed adaptively for 1 week,randomly divided into normal control group（Group A,n=10）,the rest of the model replication group.After 4 weeks of feeding,the model replication group was modeled by 50mg/kg of STZ intraperitoneal injection.Divided the successful models into DM group（Group B,n=10）,Chinese Medicine Group（Group C,n=10）.Group C was given JDTLBSS,each 4ml,4ml each day;Group A and Group B were given the 4ml distilled water.At the 8^thweekend prepare kidney homogenate.ELISA method to detect Bax,Bcl-2,Caspase-3 expression level in kidney homogenate.Results:1.Group B,C compared with Group A,fasting plasma glucose,serum creatinine,urea nitrogen and urinary microalbumin were all increased,and Group C compared with group B,fasting blood glucose,serum creatinine,urea nitrogen and urinary albumin were all decreased.2.immune histochemical method detection results show that compared with Group A,Bcl-2 lower expression,but Bax,and rate of Bax/Bcl-2 were increased significantly in group B and C;compared with Group B,Bax was depressed expression but Bcl-2 incresed expression,so the rate of Bax/Bcl-2 epression was obviously decreased in Group C.Compared with Group A,Caspase-3 was increased significantly in Group B,compared with Group B,Caspase-3 was showed lower expression in group C.Conclusion:1.Jiedu Tongluo Baoshen Method can reduce the DM rat fasting plasma glucose,protect the kidney runction,reduce the urine microalbumin.The mechanism may be lowered by the expression of Bax,the increase of Bcl-2 expression,the decrease of Bax/Bcl-2ratio,Thus,the effect of inhibiting the apoptosis of the podocyte by reducing the production of Caspase-3,and protect the glomerular filtration barrier.2.Preliminary illustrates the Jiedu Tongluo Baoshen Method protect the kidney prdocyte of experimental diabetic rats by means of regulate apoptosis mechanism such as Bax,Bcl-2,Caspase-3.
