| Diabetes is a major disease which is harmful to human health.The prevalence is rising in recent years and it is difficult to cure.People are actively involved in seeking effective drug treatment and development.Iron is an indispensable trace element in the human body,which have decisive significance for human life activities.State of iron remains relatively stable in the body and iron-deficiency can result in iron deficiency anemia,but the iron overload and oxidative stress can lead to the body appears peroxidation damage which is one of the causes of diabetes.Kiwi fruit leaf flavonoid compounds is a kind of phenolic compounds,which can alleviate oxidative damage in the body,can chelate iron in the body,reduce the oxidative damage and relieve oxidative stress,but kiwi fruit leaf flavonoids cures type 2 diabetes by reducing iron overload which is rarely reported.This study intend to the T2 DM rats induced by high-sugar and high-fat diet supplement kiwi fruit leaf flavonoids,skeletal muscle SOD,8-OHdG,CAT level change,clear flavonoid compounds in the body of rat and regulation of iron metabolism regulating fasting blood glucose,insulin sensitivity and the important role of type 2 diabetes,for type 2 diabetes drug development provides new ideas.Methods:8 rats were randomly extracted from 30 twelve-month old male SD rats as the control group(NC group,fed with common forage),the rest 22 rats were made T2 DM model(fed with high-sugar and high-fat forage for 5 weeks and injected STZ 35mg/kg)when the FBG of rats exceeded to 11.1mmol/L.And the diabetic model rats were randomly divided into diabetic control group(DC group,n=8)and supplied with Kiwi fruit leaf flavonoids group(DK group,n=8,with 40/(kg.d)Kiwi fruit leaf flavonoidsto lavage).After 5 weeks,the contents of serum ferritin and insulin were examined by ELASA.Muscle,liver MDA and muscle SOD,CAT were examined by reagent kit.To detect all the 12 h experimental rats fasting glucose with the fast blood glucose analyzer;Serum insulin,8 hydroxy deoxyguanosine,serum ferritin were detected with enzyme-linked immunosorbent assay;Glycogen,serum iron,superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT),cholinesterase level,lipid metabolism index(HDL and LDL,CEH,TC),serum ferritin and serum insulin were measured with biochemical kit;Leucocyte number,number of red blood cells were measure with medical routine blood hemoglobin concentration index(trend trade HF-3800);Rt-pcr detection of liver hepatic m RNA levels.Results:(1)The amount of blood glucose,serum insulin,HOMA-IR,in group NC and group DK were obviously lower than the diabetic control rats(DC group,P<0.01,P<0.05);The amount of HOMA-SIS,hepatic glycogen obviously higher than the diabetic control rats(DC group,P<0.01).(2)The amount of Ft,liver iron,hepatic hepcidin mRNA in group NC and group DK were obviously lower than the diabetic control rats(DC group,P<0.01,P<0.05)(3)The amount of muscle MDA,muscle 8-OHdG,liver 8-OHd G in group NC and group DK were obviously lower than diabetic control rats(DC group,P<0.01,P<0.05);The amount of muscle SOD,CAT did not change significantly between group NC and group DC.And The amount of muscle SOD and CAT in group DK were obviously higher than the diabetic control rats and normal control.(P<0.01).Conclusions:(1)Long-term high-sugar and high-fat diet can increase the blood sugar,reduce insulin sensitivity,increase insulin resistance,iron overload and oxidative damage in the rat.(2)Kiwi fruit leaf flavonoid could reduce iron reserves and liver iron deposition,but improve reductase activity and relieve oxidative damage.(3)Kiwi fruit leaf flavonoid reduce the rat’s inflammatory response,reduce Hepcidin mRNA expression,promote the release of the iron storage in the liver,reduce the liver iron storage,alleviate peroxidation damage of liver and skeletal muscle,improve insulin sensitivity and decrease insulin resistance,alleviate symptoms of T2DM. |
