Discussion On The Effect And Mechanism Of Qingre Tongluo Fang On Acute Gout Based On TLRs Pathway

Objective: To explore the effect and mechanism of Qingre Tongluo Fang from the perspective of TLRs and their signal pathways,and provide theoretica l basis for the treatment of acute gouty arthritis with Qingre Tongluo Fang.Methods: 40 patients with acute gouty arthritis were selected by random number table method.They were divided into treatment group(20 cases)and c ontrol group(20 cases).They were given Qingre Tongluo Fang(granules;1 do se/day)and colchicine.Tablets(1.5 mg/day)were treated for 1 week.By comp aring the relevant scores before and after treatment of the two groups of patie nts(pain evaluation of the patient,evaluation of the joint tenderness of the do ctor,evaluation of the patient’s health),scores of syndromes of traditional Chin ese medicine,and ESR,CRP,UA,evaluation of Qingre Tongluo Fang for acut e gout Clinical efficacy of arthritis;RT-qPCR was used to detect TLR-2,TLR-4,and IL-1βmRNA in peripheral blood mononuclear cells(PBMCs)and and El isa was used to detect IL-1β,IL-8,IL-10,IL-37 levels in serum before and af ter treatment in both groups.To investigate the effect of Toll-like receptors and related factors and reveal their mechanism of action.Results: 1.Qingre Tongluo can improve the patient’s correlation score(pat ient pain evaluation,doctor joint tenderness evaluation,patient health evaluation),TCM syndrome scores,compared with the control group(colchicine group),no significant statistical significance(P>0.05).2.Qingre Tongluo can significantly improve the patients’ biochemical detec tion indicators(ESR,CRP,UA),and reduce the CRP,UA levels better than th e control group,the results were statistically significant(P <0.05,P <0.01).3.Qingre Tongluo can reduce the levels of TLR-2,TLR-4,IL-1βmRNA a nd serum IL-1β,IL-8,IL-10 and IL-37 in PBMCs(P<0.01).In the control gro up(colchicine group),the results were statistically significant(P<0.01).Conclusion:1.Qingre Tongluo treatment of acute gout efficacy.2.The therapeutic effect of Qingre Tongluo may be related to the inhibiti on of TLRs and their signaling pathways.
