Objective: observation of the Biling tongfeng decoction on the treatment of hyperuricemia in animal experiments,and to further explore its mechanism.Methods: sixty male healthy Sprague-Dauley rats were randomly divided into blank control group(group A),model group(group B),Biling tongfeng decoction high dose group(group C),Biling tongfeng prescription middle dose group(group D),Biling tongfeng party low dose group(group E),allopurinol group(group F),n = 10.In the morning,except the blank group fed with same cubage of distilled water,the other five groups were fed with adenine(100mg/kg)plus ethambutol hydrochloride(250mg/kg)to establish the rats models of hyperuricemia.In the afternoon,except blank group and model group fed with distilled water,the Biling tongfeng decoction high dose(24g/kg),middle dose(12g/kg),low dose(6g/kg)groups and allopurinol group(10mg/kg)were respectively fed with different concentrations(1.6g/ml,0.8g/ml,0.4g/ml)of Biling tongfeng decoction drug solution and allopurinol(0.68mg/ml)according to the weight of l5ml/kg,one time a day.After 14 days of experiment,all rats were intraperitoneally injected with urethan to anesthetize,and then abdominal aortic blood collection,excised the left kidney and part of liver tissue rapidly.Blood samples were taken for detection of the level of serum uric acid,creatinine,urea nitrogen and xanthine oxidase(XO)of experimental rats;take the liver tissue for detection of liver xanthine oxidase(XO)activity;renal tissues were stained by HE pathological observation;the expression of organic anion transporter 1(OAT1),organic anion transporter 3(OAT3)protein in the renal tissue tubular epithelial cells were detected by immunohistochemical assay.Results: Biling tongfeng decoction high dose group,Biling tongfeng decoction middle dose group and allopurinol group could effectively decrease the level of experimental rats serum uric acid;the level of serum uric acid in high dose of Biling tongfeng decoction group was similar to that in allopurinol group.The high dose, medium dose of Biling tongfeng decoction groups and allopurinol group could obviously reduce the level of experimental rats serum creatinine and urea nitrogen;the level of experimental rats serum creatinine,urea nitrogen in medium dose group of Biling tongfeng decoction was similar to that in allopurinol group;the level of serum creatinine and urea nitrogen in high dose group of the Biling tongfeng decoction is obviously lower than in allopurinol group.Allopurinol group,Biling tongfeng decoction high dose group,medium dose group could obviously reduce the activity of serum and liver XO;the activity of xanthine oxidase in high dose,medium dose of Biling tongfeng decoction groups were obviously higher than in allopurinol group.The Biling tongfeng decoction high dose,middle dose group and allopurinol group could decrease deposition of urate crystal and infiltration of inflammatory cells in renal tubules and interstitial sites of experimental rats,high dose group is most obvious.The high dose of Biling tongfeng decoction groups could raise the protein expression of OAT1;The Biling tongfeng decoction high dose and middle dose group could raise the expression of OAT3 protein.Conclusion: 1 Biling tongfeng decoction can obviously reduce the levels of serum uric acid,creatinine,urea nitrogen of experimental rats,improve the function of kidney,the level of serum uric acid of high dose is similar to that in allopurinol group;the improment of kidney function of Biling tongfeng decoction high dose group is better than allopurinol group.2 Biling tongfeng decoction can improve kidney pathological morphology,protect and repair the nephron,alleviate the renal inflammatory reaction.3 Biling tongfeng decoction can decrease the activity of serum and liver XO.4 Biling tongfeng decoction can up-regulate the protein expression of OAT1 and OAT3.5 The mechanism of the Biling tongfeng decoction decrease serum uric acid level may be related to the down-regulation activity of XO,up-regulation protein expression of OAT1,OAT3. |
