Objective: Establish the HepG2 cell model infected with hepatitis C virus and hepatic steatosis.Observe the influence of Jianpi Qinghua Decoction to the content of ApoB,VLDL,TG,LEP and the quantity of HCV RNA in the HepG2 cell superntant,and elucidate the effect of Jianpi Qinghua Decoction on the anti-HCV effect of interferon and mechanism.Methods: 1.40 SPF grade SD rats were divided into four groups,10 rats in each group.Control group: Give normal saline for 10 days.Western medicine group: Pegylated interferon α-2a injection was injected subcutaneously and ribavirin gavage.Chinese medicine group: Jianpi Qinghua Decoction gavage.Combination group: Pegylated interferon α-2a injection was injected subcutaneously,ribavirin gavage and Jianpi Qinghua Decoction gavage.After 10 days,take blood from rats abdominal aorta to prepare drug containing serum.2.Establish the HepG2 cell model infected with hepatitis C virus.This group is model group,another blank control group.The cells and supernatants of 48 h,96h,144 h,192h were collected in blank group and model group respectively.3.HepG2 cells infected with HCV,which were treated with Rats contain serum in each group,and the cells and supernatants of 48 h,96h,144 h,192h were collected in each group respectively.4.ELISA method was used to detect cell supernatants of ApoB,VLDL,TG,LEP content.The HCV load was measured by RT-PCR in cells.Result:Blank group and model group ApoB,VLDL,TG,LEP content: At 96 h,144h,192 h,the ApoB content in the model group was significantly lower than that in the blank group(P<0.05).At 144 h,192h,the VLDL content in the model group was significantly lower than that in the blank group(P<0.05).At 144 h,192h,the TG content in the model group was significantly higher than that in the blank group(P<0.05).At 144 h,192h,the LEP content in the model group was significantly higher than that in the blank group(P<0.05).Blank groupand model group are compared at different times,ApoB,VLDL contents in the model group showed a decreasing trend with time(P<0.05).TG contents in the model group showed a rising trend with time(P<0.05).There was interaction between different groups and time(P<0.05).ApoB,VLDL,TG content was significantly different due to different treatment factors(P<0.05).After Intervention of Containing serum:1.ApoB content: At 96 h,compared with the control group,western medicine group,the TG content of combination group was significantly increased(P<0.05).At 144 h,compared with the control group,the content of ApoB in the combination group was significantly increased(P<0.05).Compared with the western medicine group,the content of ApoB was increased in the Chinese medicine group and the combination group,the differences were statistically significant(P<0.05).At 192 h,compared with the control group and the western medicine group,the content of ApoB in the Chinese medicine group and the combined group all increased(P<0.05).Compared with the Chinese medicine group,the content of ApoB increased significantly in combined group(P<0.05).The content of ApoB in the combination group,the western medicine group and the Chinese combination group showed a slowly declining trend with time(P<0.05).The interaction existed between different groups and time(P<0.05).There were significant differences in ApoB content of all groups because of different processing factors(P<0.05).2.VLDL content: At 144 h,compared with the control group and western medicine group,the content of VLDL in the combination group was significantly increased,the differences were statistically significant(P<0.05).At 192 h,compared with the control group and the western medicine group,the VLDL content of the Chinese medicine group and the combination group was significantly increased(P<0.05).The content of VLDL in combination group was significantly higher than that in Chinese medicine group(P<0.05).The content of VLDL in the combination group,the western medicine group and the Chinese combination group a slowly declining trend with time(P<0.05).The interaction existed between different groups and time(P<0.05).There were significant differences in VLDL content of all groups because of different processing factors(P<0.05).3.TG content: At 48 h,compared with the control group,the TG content of the combination group decreased significantly(P<0.05).Compared with the western medicine group,the TG content of the Chinese medicine group and the combination group all decreased(P<0.05).Compared with the Chinese medicine group,the TG content of the combined group was lower(P<0.05).At 96 h,compared with the control group,the TGcontent in the western medicine group,the Chinese medicine group and the combination group decreased significantly(P<0.05).Compared with the western medicine group and the Chinese medicine group,the TG content of the combined group decreased significantly(P<0.05).At 144 h,192h,compared with the control group,the TG contents of the western medicine group,the Chinese medicine group and the combined group were significantly decreased(P<0.05).Compared with the western medicine group,the TG content of the Chinese medicine group and the combined group decreased significantly(P<0.05).Compared with the Chinese medicine group,the TG content of the combined group decreased significantly(P<0.05).The content of TG in the combination group,the western medicine group and the Chinese combination group a slowly rising trend with time(P<0.05).The interaction existed between different groups and time(P<0.05).There were significant differences in TG content of all groups because of different processing factors(P<0.05).4.LEP content: At 96 h,the LEP content of the combined group was significantly lower than that of the control group(P<0.05).At 144 h,compared with the control group,the LEP content of the traditional Chinese medicine group and the combined group decreased(P<0.05).Compared with the western medicine group,the combined group was significantly decreased(P<0.05).At 192 h,compared with the control group,the levels of LEP in the Chinese medicine group and the combined group were significantly lower than those in the control group(P<0.05).Compared with the western medicine group,the levels of LEP in the traditional Chinese medicine group and the combined group were significantly lower than those in the control group(P<0.05).Compared with the Chinese medicine group,LEP content in the combined group was significantly decreased(P<0.05).There were significant differences in LEP content of all groups because of different processing factors(P<0.05).5.Viral quantity: At 96 h,compared with the control group,the HCV-RNA quantity of the western medicine group and the combined group decreased(P<0.05).Compared with the Chinese medicine group,the HCV-RNA quantity of the combined group decreased significantly(P<0.05).At 144 h,192h,compared with the control group,the HCV-RNA quantity of the western medicine group and the combined group decreased significantly(P<0.05).Compared with the western medicine group,the HCV-RNA quantity of the combination group decreased significantly(P<0.05).Compared with the Chinese medicine group,the HCV-RNA quantity of the combination group decreased significantly(P<0.05).The quantity of HCV-RNA in the combination group and the western medicine group show a slowly rising trend with time(P<0.05).The interaction existed betweendifferent groups and time(P<0.05).There were significant differences in the quantity of HCV-RNA between the two groups because of different processing factors(P<0.05).Conclusion:1.HCV infection reduce the content of ApoB and VLDL,increase the content of TG and LEP in the supernatant of HepG2 cells.It is suggested that the virus infection may interfer the metabolism of lipid.2.Jianpi Qinghua Decoction inhibit the ApoB,VLDL content of the continuous reduction,and inhibit the TG,LEP content of the continuous increase.It suggested that Spleen Qinghua Decoction could regulate the contents of lipid in supernatant of HepG2 cells infected with HCV.It has a up-regulation of ApoB and VLDL content.It has a down-regulation of TG and LEP content.3.The combination of Jianpi Qinghua Decoction,interferon and riboflavin could not only affect ApoB,VLDL,TG and LEP metabolism in supernatant of HepG2 cells,but also had significant inhibitory effect on hepatitis C virus.The results showed that Jianpi Qinghua Decoction can enhance the anti-viral ability of interferon,its mechanism may be related to the improvement of lipid metabolism... |
