This topic aims to study the new formulations of insulin-quick-acting soluble insulin polymer microneedles.The microneedle matrix was digested with Oligo-sodium hyaluronate,and the drug site was sublingual and the microneedle array was 225(15x15)per square centimeter.In order to prevent the drug from dissolving and dispersing in the oral cavity,a blended solution of PVA and CMC-Na was used as the backing layer.Establishment of type 2 diabetes model: intraperitoneal injection of STZ induced selective injury of islet β cells in male SD rats.Mainly on the hardness of the microneedles,the microneedles to examine the hardness of the aluminum foil by rolling.The microneedles were placed under the tongue of the rats and observed every 5 minutes to record the dissolution time of the microneedles in the mouth.Taken 1 cm2 microneedles dissolved in 0.01 mol/L hydrochloric acid and diluted to 10 m L,measured at 214 nm peak area,into the standard curve to calculate the drug content;by giving Type 2 diabetes mellitus,the effects of microinjection of insulin on the onset time and hypoglycemic effect were studied.Insulin sublingual soluble polymer microsicles had hypoglycemic effect on diabetic rats.The application prospect of insulin-sensitive micro-sublingual patch administration system in insulin non-injection dosage form is promising.
