Objectives:To analyze the chromosome karyotype of the couples undergoing IVF /ICSI-ET,and discuss the necessary of chromosome analysis before assisted reproductive technology.Provide guidance for genetic counseling and the choice of assisted reproductive method.Methods:2 621 patients undergoing IVF/ICSI-ET in Reproductive Medicine Center Hospital of Tianjin Medical University during 1999.1 to 2015.12 were taken in the study,including 1 365 males and 1 256 females.A total of 1 247 couples have taken chromosome karyotype examination.The gender,the age,the number of abortions as well as semen condition are checked in medical records,using the method of G-banding karyotype analysis of the peripheral blood,analyze the rate of abnormal chromosome karyotype of the couples undergoing IVF/ICSI-ET.Taking the abnormality rate and polymorphism rate in general population as control.The results were analyzed using SPSS17.0 software,P < 0.05 was statistically significant.Result:1.71 cases chromosome abnormality were found in the 2 621 cases with the abnormal rate of 2.71%,the rate of general population was 0.5%.The difference between the IVF/ICSI-ET couples and the general population was statistically significant(P<0.0001).2.35 cases of chromosome abnormality were found in the1 365 male patiens with the abnormal rate of 2.56%.36 cases of chromosome abnormality were found in the1 256 females patients with the abnormal rate was 2.87%.The chromosome abnormality rate in male and female in this study were higher than in the general population((P<0.0001),The difference of the chromosome abnormality rate between the men and the women have no statistically significant(P=0.634).3.In the 1 365 male patiens,3 cases of chromosome abnormality were found in the 527 cases with normal sperm condition,the abnormal rate was 0.57%,compared with the general population,the difference have no statistically significant(P=1).14 cases of chromosome abnormalities were found in the 580 males with mild oligozoospermia,the abnormal rate was 2.41%;6 cases of chromosome abnormalities were found in the 239 with severe oligozoospermia,the abnormal rate was 11.30%;12 cases of chromosome abnormalities were found in the 19 males with non-obstructive azoospermia,the abnormal rate was 63.16%.The rate of chromosome abnormalities in the patients with abnormal sperm condition is higher than in the general population,The difference have statistically significant(P<0.001).4.21 couples have checked with chromosome abnormality in the 371 couple with the history of one time abortion,the rate was 5.36%.24 couples were found with chromosome abnormality in the 423 couples with the history of two more times spontaneous abortion,the rate was 5.56%;The difference have no statistically significan(P=0.9).5.127 cases of chromosome polymorphism was detected among the 2 621 case patients,the chromosome polymorphism rate was 4.85%,comparing with the rate of chromosome polymorphism in the general population,the difference was statistically significant(P<0.0001).82 cases of chromosome polymorphism were found in the 1 365 males,the polymorphism rate was 6.01%;45 cases of chromosome polymorphism were found in the 1 256 females,the polymorphism rate was 3.58%.The difference of between the males and females was statistically significant(P=0.004).9 cases chromosome polymorphism in the 527 cases with normal sperm condition,the chromosome polymorphism rate was 1.71%,The difference of polymorphisml rate between patients with normal sperm condition and the general population was no statistically significant(P=0.916).41 cases chromosome polymorphism were detected in the 580 cases with mild oligozoospermia,the rate was 7.07%;27 cases of chromosome polymorphism were found in the 239 cases with severe oligozoospermia,the rate was 11.30%.5cases of chromosome polymorphismin were found in 19 males with non-obstructive azoospermia,the polymorphism rate was 26.32%.The difference have statistically significant(P<0.0001).Conclusion:1.The abnormal rate of chromosome between the IVF/ICSI-ET couples and the general population is different with statistical significance.2.The abnormal rate of chromosome in males undergoing the IVF/ICSI-ET with abnormal sperm condition is statistically significant different from the general population.The difference of abnormal rate of chromosome between males with normal sperm condition and the general population was no statistically significant.3.The times of abortion have no relation with the chromosome abnormality in the couples4.The difference of polymorphism rate of chromosome between the IVF/ICSI-ET couples and the general population was statistically significant.5.Chromosome analysis before undergoing IVF /ICSI-ET was necessary,It can make clear that whether the chromosome is normal and help to make choice of which methods to choose in the assisted reproduction technology.,which not only increase the success rate of IVF /ICSI-ET,but also decrease the birth of chromosomal disease.
