| Objective:(1)Detecting the expressions of tissue factor positive microparticles in plasma of patients with malignant solid tumors,so as to investigate the clinical significance of the expressions of TF+MP in malignant solid tumors.(2)Comparing the expressions of TF+MP in different stages of malignant solid tumors,and to investigate whether it is associated with tumor’s progression.(3)Investigating the expressions of TF+MP in malignant solid tumors with thrombosis,and to explore the role of TF+MP in tumors with thrombosis.(4)Investigating the changes of TF+MP expressions in patients with malignant solid tumors before and after anesthesia,and to explore the influence of the anesthetic factor on TF+MP in tumor patients.(5)Investigating the changes of TF+MP expression in patients with malignant solid tumors before operation and after operation,and to explore the possible causes of thrombosis after operation of malignant solid tumor.Methods:36 cases of malignant solid tumor patients with surgery as the experimental group(n=36)in our hospital and 29 cases of healthy people as the control group(n=29).Comparison of TF+MP changes in patients with malignant solid tumor and comparing the expressions of TF+MP in the different stages,with thrombosis,the pre-anesthesia and post-anesthesia and the pre-operation and post-operation: The control group was taken in the early morning fasting venous blood,and the experimental group was given the fasting venous blood before anesthesia,after anesthesia and after operation.After two times centrifugation,the platelet poor plasma was obtained.The expression levels of TF+MP in plasma of experimental group and control group was detected by immunofluorescence combined with flow cytometry.And then get the target particle TF+MP: diameter 0.1-1.0ul,FITC-Annexin-V and PE-TF antibody double positive.Finally,the expressions of TF+MP in all groups were compared.Results:(1)The expression levels of TF+MP in patients with malignant solid tumor and control group: The expression of TF+MP in malignant solid tumor patients was higher than that in healthy group(P<0.001).It is suggested that malignant solid tumor can promote the expression of TF+MP.(2)The expression levels of TF+MP in different stages of tumor: The level of TF+MP in patients with stage I cancer was not significantly different from that in healthy group(P=0.261).The levels of TF+MP in stage from II to IV tumors was significantly higher than that in stage I cancer(P<0.05).However,there was no significant change in the expression levels of TF+MP in stage II,III and IV tumors(P>0.05).It is suggested that the expression of TF+MP in malignant solid tumors in advanced stage is significantly higher.(3)The expression level of TF+MP in tumor with VTE: The expression of TF+MP in tumor patients with VTE was significantly higher than that in patients without VTE: 33%.It is suggested that TF+MP may play an important role in the thrombosis of malignant solid tumors.(4)The expression levels of TF+MP in patients with malignant solid tumor before and after anesthesia: There was no obvious change in the expressions of TF+MP in the patients with malignant solid tumor before and after anesthesia(P=0.838).It is suggested the anesthetic factor on postoperative TF+MP expression did not have a significant impact.(5)The expression levels of TF+MP in patients with malignant solid tumor before and after operation: The expressions of TF+MP was increased in patients with malignant solid tumors after surgery(P=0.042).It is suggested that surgery could promote the release or expression of TF+MP.Conclusions:(1)The expression levels of TF+MP in peripheral blood of patients with malignant solid tumor was higher than that in healthy control group.And the levels of TF+MP in patients with stage II,III and IV were significantly higher than those of patients with stage I.It is suggested that TF+MP is highly expressed in malignant solid tumors,and the expression level of TF+MP is increased with the progression of tumor,which may indicate the occurrence of malignant solid tumors and reflect the progress of tumor.(2)The expression levels of TF+MP may be closely related to the thrombosis of malignant solid tumors.(3)The high expressions of TF+MP may be associated with the thrombosis of malignant solid tumors after operation.And the anesthetic factor does not affect the expression of TF+MP in tumors,which is not related to the thrombosis of the malignant solid tumor after operation.In conclusion,TF+MP may be associated with the occurrence,progression and thrombosis of malignant solid tumors,and may be an important factor in the occurrence of thrombosis in patients with malignant solid tumors after surgery. |
