Objective:Our study aimed to discuss the impact of FBXW7 gene point mutation on cell proliferation,apoptosis,migration,and invasion processes of colorectal cancer cell strain HCT-116.Methods:Recombinant plasmids were constructed by having wild-type and mutant-type FBXW7 SNP overexpressed respectively;the transfected HCT-116 strains were built by having the plasmids transfected into the vector cells;and then the FBXW7 protein expression level of transfected HCT-116 strains was detected by Western blotting.Subsequently,cell proliferation capacity was tested by CCK-8 assay;tumor cell clonality was tested by HTCA;cell apoptosis function was tested by FCM;cell migration and invasion were tested by checking scratches and Transwell.Results:Higher HBXW7 protein expression level was detected in transfected wild-type HCT-116 strains in comparison to the levels of the placebo-control(the uninfected strains),negative-control(strains transfected by empty plasmids),and control(strains transfected by the mutant-type FBXW7 plasmids)(P<0.05).Compared with the other groups,strains transinfected by the wild-type HBXW7 exhibited significantly reduced proliferation,clonality,migration and invasion rate,and lifted apoptosis rate.Conclusion:FBXW7 gene point mutation can affect protein expression of FBXW7,by which result in incapacity of FBXW7 gene on suppression of cell proliferation,migration and invasion,as well as promotion of apoptosis in the HCT-116 strains.
