OBJECTIVE: To observe the inliuence of liver cells and liver Sox9 、 Ngn3 、 Hes1 expression condition in the early stage of non obese diabetic mice which interfered by Adenovirus vector-mediated murine interleukin and Anti CD20 monoclonal antibody.METHOD: 24 NOD mice,17-19 weeks,female,were randomly divided into(A)group Anti-CD20,(B)group Anti-CD20 + IL-10,(C)group IL-10,(D)group physiological saline(Normal control,NC).Injected Anti-CD20,Anti-CD20 + IL-10,IL-10 and physiological saline on day 1,day 3,day6 and day 9 via tail vein.Monitored blood glucose weekly,detected serum Insulin,IL-10 and CD20 level after 9 weeks.Monitored the expression of IL–10,CD20,Insulin and Sox9 、 Ngn3 、 Hes1 of liver cells in immunohistochemical method.RESULTS AND CONCLUSION : Association of Anti-CD20 and IL-10 can control the blood glucose of NOD mice effectively,can increase the serum insulin,IL-10 and CD20 expression level,and can promote the expression of insulin,IL-10 in liver cells.Combined intervention can reduce the levels of inflammatory factors such as IL-17 and IL-1 β,promote the synthesis of TGF-β,and maintain immune homeostasis.And,the combined intervention can activate the Notch signaling pathway,promoting Sox9,Ngn3 gene expression to promote the synthesis of liver IPS(islet endocrine cells),hepatic insulin secretion,has a protective effect on survival of islet beta cell function. |
