| Objective To explore the relationship between the expression of CUGBP1 and the biological characteristics of lung cancer cells.Methods Lung cancer cell lines were subcultured and human lung adenocarcinoma cell line PC14 was selected as an experimental object and cultured using enzymic digestion and subculture method.Expression vectors of lentivirus CUGBP1-sh RNA and CUGBP1-c DNA were constructed and transfected into PC14 lung cancer cell lines,respectively.As a control variable,the expression of CUGBP1 was up-and down-regulated.The expression of CUGBP1 of each cell line was detected by using RT-PCR and Western Blot methods.The effects of the mutual regulation of factors on chemotaxis and invasion of lung cancer cells were studied by cell invasion assay(basement membrane invasion assay),scratch test,Transwell,and adhesion test to further explore the relationship between CUGBP1 expression and the biological characteristics of lung cancer cells at the cytological level.Results The mRNA and protein expression of CUGBP1 in patients with lung cancer is associated with TNM stage and differentiation,and therefore is an independent prognosticfactorof patients with lung cancer.The effect of CUGBP1 is a complex phenomenon resulted from multi-gene synergy.Conclusion The over-expressed CUGBP1 regulates the CXCL1/2-S100A8/9 paracrine pathway and is involved in the biological response of lung cancer cells,providing a new breakthrough point for the targeted therapy of lung cancer. |
