Objective: The effect of the new addition of New Yellow Dragon Decoction(NYDD)on the adhesive intestinal obstruction(AIO)in experimental rats and the observation of the results were analyzed.The effect and mechanism of NYDD on AIO were studied.The clinical application and teaching of Chinese medicine provide experimental basis.Method: 32 male SD purebred healthy rats,weight 200 ± 20 g were fed for one week.According to random grouping method is divided into four groups: Normal control group(N),Model group(M),New Yellow Dragon Decoction group(NYDD)and Colle Seed Oil group(CSO).Each group of eight rats.In addition to the blank group,the remaining groups were modeled.In this study,the animal model was established by Clamping trauma method to establish an adhesive intestinal obstruction model combined with 7 thin line ligation of the ileocecal method.Establishment of rat model of adhesive intestinal obstruction.After the sub-cage feeding,recovery of drinking water and feeding,observe the general situation of rats,and measure the weight.The next day after the operation,new yellow dragon decoction group given the new yellow dragon decoction 2ml / 100g(including crude drug dose 7.2g / kg),seed oil group given 1ml / 100g(containing crude drug dose 3.2g / kg),The remaining two groups were given the same amount of saline treatment.On the seventh day after surgery,abdominal aorta blood was taken,blood routine,plasma D-lactate levels,serum endothelin(ET)were measured.Take 1cm intestinal tissue.HE stains was used to observe the pathological changes of intestinal tissue and statistically analyzed.result:1 The general condition of the ratsThere was no significant change in the mental state,activity,hair,diet,defecation and so on in the control group.The rats in the model group were in a state of malaise,decreased activity,unresponsive,bleak color,significantly reduced eating,and a significant decrease in body weight.Modeling the third day,Compared with the control group,There was a significant difference in the reduction in food intake between the model groups(P < 0.01),There was no difference in weight loss(P > 0.05).Compared with the model group,There was no difference in the intake and weight loss between new yellow dragon decoction group and seed oil group(P > 0.05).Compared with new yellow dragon decoction group and seed oil group,There was no statistically significant difference in food intake and weight loss(P > 0.05).Modeling the fifth day,Compared with the control group,There was a significant difference in the reduction in feed intake and weight loss in the model group(P < 0.01).Compared with the model group,there was a difference in the increase in the amount of food intake and the decrease in body weight in the new yellow dragon decoction group(P < 0.05),there was no difference in the increase in food intake and the decrease in weight loss(P < 0.05).Compared with the new yellow dragon decoction group and the seed oil,there was no difference in the increase in food intake and the decrease in weight loss(P < 0.05).Modeling the seventh day,compared with the control group,the rats in the model group had significant changes in both food intake and weight loss(P < 0.05).Compared with the model group,there was no significant difference between the new yellow dragon decoction group and the seed oil in the increase of food intake and the decrease of body weight(P < 0.05).Compared with the new yellow dragon decoction group and the seed oil group,there is a difference in the increase in food intake(P < 0.05),there was no significant difference in weight loss(P < 0.05).2 Observation of serum D-lactic acid and serum endothelin(ET)D-lactic acid content determination results :Compared with the control group,the content of D-lactic acid in the model group was significantly increased,the difference was statistically significant(P < 0.05).Compared with the model group,the decrease of D-lactic acid content in the new yellow dragon decoction group and the seed oil group was statistically significant(P < 0.05).Compared with the new yellow dragon decoction group and the seed oil group,D-lactic acid content was statistically significant(P < 0.05).ET measurement results: Compared with the control group,the ET level in the model group was significantly increased(P < 0.01).Compared with the model group,the content of ET in the new yellow dragon decoction group was decreased,the difference was statistically significant(P < 0.05).There was no significant difference in ET content between the seed oil group(P < 0.05).Compared with the new yellow dragon decoction group and the seed oil group,there was a difference in ET reduction in rats(P < 0.05).Blood routine results: Compared with the control group,the number of white blood cells and neutrophils in the model group increased significantly,the difference was statistically significant(P < 0.05).Compared with the model group,there was significant difference in the number of white blood cells and neutrophils in the new yellow dragon decoction group(P < 0.01).There was a statistically significant difference in leukopenia between the rapeseed oil group(P < 0.01).There was a significant difference in neutropenia(P < 0.05).Compared with the new yellow dragon decoction group and the seed oil group,there was significant difference in leukopenia(P < 0.01),and there was a significant difference in the number of neutrophils(P < 0.05).Platelet assay results: Compared with the control group,the platelet count of the model group(P < 0.01).Compared with the model group,there was significant difference in platelet count between the new yellow dragon decoction group and the seed oil group(P < 0.01).Compared with the new yellow dragon decoction group and the seed oil group,Platelet count reductions are different(P < 0.05).3 Blood viscosityCompared with the control group,there was significant difference in the whole blood viscosity and whole blood viscosity of the model group at different shear rates(P < 0.01),there was no significant difference in plasma viscosity and hematocrit(P > 0.05).Compared with the model group,there were significant differences in whole blood viscosity and reduced viscosity between the new yellow dragon decoction group and the seed oil group at different shear rates(P < 0.01),there was no significant difference in plasma viscosity and hematocrit(P > 0.05).Compared with the new yellow dragon decoction group and the seed oil group,there was a difference in whole blood viscosity,whole blood reduction viscosity,and plasma viscosity(P < 0.05).There was no difference in hematocrit(P > 0.05).4 Pathological observation of ileumCompared with the control group,the lymphoid cells of the model control group had a lot of lymphocytes,the villus epithelial cell structure was damaged,the inflammatory cells were infiltrated,the inflammatory cells,disappearance of goblet cells,swelling,necrosis and off.At the same time,interstitial obvious edema,cell boundaries are unclear,a lot of inflammatory cells appear.Compared with the model control group,the swelling of the ileocecal epithelium in the new yellow dragon decoction group rats was significantly reduced,the infiltration of inflammatory cells was significantly reduced,and the edema and exudation were not obvious.Seed oil group with inflammatory edema,epithelial cells found necrosis and shedding.conclusion:1 Through the observation of the general situation of experimental AIO rats,new yellow dragon decoction can promote AIO rats to increase the amount of defecation,reduce weight loss.Indicating that can improve the general state of experimental obstruction rats,confirmed its effect of Yiqifuzheng and Tongfu.2 New yellow dragon decoction can reduce serum ET,D-lactic acid levels.The results showed that the new yellow dragon decoction could slow down the development of intestinal obstruction and the degree of intestinal ischemia-reperfusion injury in AIO rats.3 New yellow dragon decoction can significantly reduce the plasma viscosity of AIO rats,and inhibit platelet aggregation.Indicating that it can strengthen the blood circulation velocity,inhibition of platelet aggregation,reduce blood flow resistance,to prevent the occurrence of AIO.4 Through the observation of the blood routine and histopathological of experimental rats,New yellow dragon decoction can reduce white blood cells,neutrophils,so that intestinal inflammatory cell infiltration decreased tissue edema.It not only relieves the role of local inflammation,but also to prevent the body’s inflammatory.5 New yellow dragon decoction and seed oil group are consistent in the defecation function,reduce the concentration of decline,less inflammatory response and no side effects.But the New yellow dragon decoction also to reduce blood viscosity,improve microcirculation,reduce inflammation,protect the role of intestinal mucosa.Proved the traditional Chinese medicine in the clinical treatment of the overall advantages,and for the clinical treatment of adhesive intestinal obstruction provides a reliable experimental basis.
