Dibaizhixue capsule(DBZX)is a traditional Chinese medicine,composed with radix rehmanniae,cacumen biotae and multiple herbs.It can be developed to cure the hemorrhagic disease caused by deficiency of yin and chronic idiopathic thrombocytopenic purpura.This study focus on its hemostasis function and anti-thrombocytopenia effect for make a correct evaluation.Objective:To observe the therapeutic effect of DBZX capsule on bleeding and coagulation disorders caused by different drugs.Methods:1 Effect of Dibaizhixue capsule(DBZX)on thrombocyt-openia model induced by anticancer drugs in mice.Sixty KM mice were divided randomly into six groups,10 numbers in each group,the control group,the model control group,the positive drug group were administrated with Shengxuexiaoban capsule(SXXB)1.4 mg/kg,and the low dosage of DBZX group 0.6 g/kg,the medium dosage group 1.2 g/kg and the high dosage group 2.4 g/kg.In addition to the blank control group,other groups were injected with cyclophosphamide 100 mg/kg intraperitoneally for 3 days.Starting from the first day of modeling drug treatment,the control and the model group were given equal volume of 0.5%sodium carboxymethyl cellulose(CMC-Na).The drugs were administrated by intragastric way each group,the volume of gavage was 20 ml/kg for 14 days.After 2hs of the last administration,clotting time and hemostasis time were measured by capillary method and cutting-tail method,platelet count(PLT)was measured by hematometer for the evaluation the hemostatic effect of DBZX.A cytarabine-induced thrombocytopenia model was modeled by intraperitoneal injection of cytarabine 50 mg/kg for 5 consecutive days.The grouping,administration and observation were the same as above.2 Effect of DBZX Capsule on warfarin-induced coagulation abnormalities in ratsForty eight SD rats were divided randomly into six groups,8 numbers in each group,the control group,the model control group,the positive drug group were administrated with Acetomenadione 2mg/kg,and the low dosage of DBZX group was 0.3 g/kg,the medium dosage group was 0.6 g/kg and the high dosage group was 1.2 g/kg.In addition to the blank control group,other groups were given to warfarin 5 mg/kg for 5 days once a day to prepare rat model of coagulation disorder.Starting from the first day of modeling drug treatment,the control and the model group were given equal volume of 0.5%sodium carboxymethyl cellulose(CMC-Na).Each group were given intragastric administration,the volume of gavage was 1 ml/100g for 14 days.After 2hs of the last administration clotting time and hemostasis time were measured by capillary method and cutting-tail method,blood platelet count(PLT)、prothrombin time(PT)、activated partial thromboplastin time(APTT)、plasma thrombin time(TT)to evaluate the hemostatic effect of DBZX.3 Effect of DBZX on the model mice with idiopathic thrombocytopenic purpura(ITP)induced by antiplatelet sera(APS).Sixty Balb/C mice were divided randomly into six groups,10 numbers in each group,the rats in the control group,the model control group,the positive drug group were administrated by prednisone 0.01g/kg,and the low dosage of DBZX group was 0.6 g/kg,the medium dosage group was 1.2 g/kg and the high dosage group was 2.4 g/kg.In addition to the blank control group,other groups of mice were intraperitoneally injected with APS,once every other day,for 9 days(1,3,5,7,9 days)for the preparation of ITP model.At the same time,daily intragastric administration,volume of 100mg/kg body weight,the blank control group and the model group were given equal volume of solvent for 10 days.At the 11th day,boold was collected from orbital venous plexus for peripheral blood cell counting.Both thymus and spleen were isolated and weighed,the organ index was calculated to observe the effect of DBZX on immune thrombocytopenia.Results:1 Effect of Dibaizhixue capsule(DBZX)on thrombocytopenia model mice induced by anticancer drugs.Compared with the model group,the middle and high dosage(1.2 g/kg,2.4 g/kg)of DBZX can significantly ameliorate the abnormality in bleeding and clotting time parameters(P<0.05 or P<0.01),and significantly increase chemotherapy-induced reduction of PLT in mice(P<0.05);2 Effect of DBZX Capsule on warfarin-induced coagulation abnormalities in rats.Compared with the model group,the middle and high dosage(0.6 g/kg,1.2 g/kg)of DBZX can significantly ameliorate the abnormality in bleeding and clotting time parameters(P<0.05 or P<0.01),significantly reduce the PT,APTT,TT(P<0.05,0.01);3 Effect of DBZX on the idiopathic thrombocytopenic purpura induced by GP-APS.Compared with the model group,the middle and high dosage(1.2g/kg,2.4g/kg)of DBZX can significantly increase platelets of ITP mice(P<0.05,0.01).The high dosage(2.4 g/kg)of DBZX significantly reduced the index of thymus and spleen of ITP mice(P<0.05).Conclusion:DBZX(1.2 g/kg and 2.4 g/kg dosage)can improve the thrombocytopenia,coagulation disorder and APS-induced thrombocytopenic purpura.DBZX can effectively improve the count of platelet,ameliorate the abnormality in PT、APTT、TT and reduce the index of thymus and spleen.The data shows that traditional Chinese medicine Dibaizhixue capsule has hemostatic effect.
