Background and objective:In recent decades,with the development of social economy and human lifestyle change,obesity rate showed a trend of rising in the world.Obesity is a chronic disease,increased the number as well as the increased volume of adipose cells are the two major pathological states of it.The body weight of the patients are more than 20% of the standard or body mass index> 25kg/m2.Obesity is an important risk factor of social mental obstacle and chronic diseases such as cardiovascular diseases,cancer,diabetes,osteoarthritis and so on.It was regarded as one of the top ten diseases that threaten human health by the World Health Organization.Therefore,it merits close attention.Embelin is a quinone compound extracted from the plant of Myrsinaceae.Studies have been shown its preventive effect on high fat diet-induced obesity and the associated complications.The aim of this study was to investigate whether Embelin ‘s effect is achieved by inhibiting lipogenesis.Methods:1.In vitro one marrow stromal cell line ST2 and mesenchymal stem cell line C3H10T1/2 were treated with dimethylsulfoxide(DMSO)and different concentrations of Embelin,oil red O staining was used to evaluate the cell differentiation degree,q RT-PCR and Western blotting were used to evaluate the lipogenic marker gene expression and its asscolated protein expression;2.Four-week-old male C57BL/6 mouse was used to construct a high-fat-diet induced obesity model,different doses of Embelin were injected subcutaneously,the body weight of each group was recorded during modeling,the mice in the experimental group and the control group were killed when the model successfully established,isolated the inguinal fat pads and recorded its weight;3.Isolated mice epididymal adipose tissue from both experimental group and control group,the expression levels of lipogenic key transcription factors and marker genes in adipose tissue were detected by q RT-PCR;4.Oral glucose tolerance test was performed in each group to determine whether embelin could improve obesity-related impaired glucose tolerance,5.A HOMA-IR analyse was carried out to determine whether embelin can improve obesity-related insulin resistance.Results: 1.Oil red O staining showed that Embelin inhibited the lipogense in bone marrow stromal cell line ST2 and mesenchymal stem cell line C3H10T1/2,the results of q RT-PCR and Western blotting showed that Embelin could inhibit the expression of sterol regulatory element-binding protein-1(Srebp-1),acetyl-Co A carboxylase 1,Acc1),fatty acid synthase(Fasn)and stearoyl-Co A desaturase(Scd1),and it’s a dose-dependent relationship;2.The body weight of mice with high fat diet was significantly higher than that of the control group,thus means the model was successfully established,While the body weight of the high fat diet group with the embelin subcutaneous injection was significantly lower than the same diet group with solvent injection at the same time,the weight of the groin fat pad in the high fat diet group was significantly higher than the same diet group with the subcutaneous injection of embelin,and it’s a dose-dependent relationship,indicating that embelin can inhibit mouse obesity;3.The expression of srebp-1,Acc1,Fasn and Scd1 in the embelin injected group was lower than that in the model group,indicating that Embelin could inhibit the lipogennesis;4.Blood glucose tolerance test in each group showed that glucose tolerance in the embelin group was stronger than that in the model group,indicating that embelin could improve obesity-related impaired glucose tolerance;5.Analysis of the Homeostasis model assessment index in each group showed that the model group index was greater than the embelin group,indicating that embelin can improve insulin resistance in obese mice.Conclusion:1.Embelin inhibits lipogenesis in ST2 cell and C3H10T1/2 cells;2.Embelin can inhibit lipogenesis and thus prevent obesity;Embelin can also improve obesity-related impaired glucose tolerance and insulin resistance.
