With the development of coastal transportation,international maritime operations,marine tourism,marine aquaculture,fishing,fishing or increasing tourism activities,as well as a wide range of military training in sea,the risk of seawater drowning personnel in our country is increasing.our country seawater drowning number is increasing year by year.Compared with freshwater,lung’s direct and secondary injury in seawater drowning is more severe because of the special physical and chemical properties of seawater such as hypertonicity,which will ultimately develop into Seawater respiratory distress syndrome(SW-RDS).SW-RDS is serious,the disease progresses rapidly.The current consensus of the treatment inlude programetiological treatment,symptomatic support treatment and mechanical ventilation.The mortality of SW-RDS is still in a high level even given positive rescue.We try to do an intervene experiment by using BMSCs and LV-CXCR4-BMSCs.Objective: To research the therapeutic potential of BMSCs and LV-CXCR4-BMSCs in treating SW-RDS.Methods: 100 SD rats were randomly divided into 4 groups(n=25),SW-RDS control group(0.5ml PBS injection in tail intravenous)、BMSCs group(2×106/ml,0.5ml BMSCs injection in tail intravenous)、LV-Con-BMSCs group(2×106/ml,0.5ml LV-Con-BMSCs injection in tail intravenous)、LV-CXCR4-BMSCs group(2×106/ml,0.5ml LV-CXCR4-BMSCs injection in tail intravenous).Killed 5 rats to take specimens at the time point 6h、1d、2d、3d、7d after the model established.Measured the serum level of TNF-α,8-iso-PGF2α and observed the right lung posterior lobe’s W/D ratio,MPO and lung permeability index.Another 6 rats were randomly divided into 2 groups(n=3),control group(GFP-BMSCs injection in tail intravenous)、experimental group(GFP-BMSCs injection in tail intravenous after the model established).Observe the engraftment situation of BMSCs in lung tissue at 6h time point after the model established.Results: 1 The interventions from BMSCs、LV-Con-BMSCs、LV-CXCR4-BMSCs to lung tissue resulted in the W/D ratio and MPO have significantly statistical differences(P<0.05)in 1d、2d、3d after the model established.What’s more,the intervention effect of LV-CXCR4-BMSCs group is better than other two groups.2 The interventions from BMSCs、LV-Con-BMSCs、LV-CXCR4-BMSCs,the serum level of TNF-α and 8-iso-PGF2α in above-mentioned time points have significantly statistical differences(P<0.05).What’s more,the intervention effect of LV-CXCR4-BMSCs group is better than other two groups.3 The lung permeability indexes of these groups only have statistical differences in the time point 7d after the model established.Conclusion: The transplantation of BMSCs and LV-CXCR4-BMSCs can reduce the degree of lung injury in SW-RDS,and the latter group has better effects. |
