| ObjectiveTo explore the effect and mechanism of granulocyte colony-stimulating factor(G-CSF)(improvement or aggravation)in rabbit models ofemphysema.Methods1 32 adult Male New Zealand white rabbits were randomly divided into 2 groups with 12 in healthy control,and 20 in emphysema model group.Choose 12 from emphysema model group randomly,the other 8 were fed for supplement.The emphysema model group was given atomized elastase(the concentraion is about 1.3%),the healthy controls took atomized normal saline(NS)for control.The models of emphysema were constructed successfully after 4 weeks,then fed normally for 6 weeks.To certify the models with comparison of peripheral blood white cells,neutrophils percentage,oxygen saturation,heart rate,lung volume,lung gross appearance and pathology between the two groups.2 The 12 of emphysema model group were divided into four groups of A,B,C,D,each group had 3 rabbits.D group acted as blank control,A,B and C acted as experimental group.Define every 4 weeks as one course of treatment and observe oxygen saturation,heart rate and lung volume to estimate that the emphysema was improved or worsen.The first course A,B and C were given G-CSF by hypodermic injection(5ug/kg qd in the first and second day,10ug/kg qd in the third,fourth and fifth day),A and B were given the second course with the same scheme,A,C(b)and D(b)were injected with antioxidant(dexamethasone 0.3mg/kg,reduced glutathione 40mg/kg,intramuscular injection,once a day for 7days).B(c)and C(c)were injected with the same antioxidant in third course.Then the emphysema model group was observed for 4 weeks with no management.Finally,all rabbits were made into pathological section,then calculate mean lining interval(MLI)and pulmonary alveoli proportion(PAP)to estimate the emphysema degree of every number of courses.Results1 To establish rabbit models ofemphysema:Peripheral white blood cells[(10.28±2.42)X109/L] and neutrophilic granulocyte percent[(43.74±8.51)%] of emphysema model group were more than healthy controls[(10.14±4.12)X109/L,(37.15±9.92)%],but there was no statistical significance(P>0.05).After the model,Oxygen saturation was significantly lower,and heart rate was significantly higer[(96.12±0.72)%,(248.33±17.52)beats per minute] compared with healthy controls[(99.00±1.35)%,(235.00±26.57)beats per minute]and before the model[(98.83±1.34)%,(230.00±12.68)beats per minute](P<0.01).After the model,lung volume[(55.62±10.00)cm3]was significantly bigger compared with healthy controls[(41.53±8.26)cm3]and before the model[(40.50±7.93)cm3](P<0.01),but there was no statistical significance between healthy controls and before the model(P>0.05).General observation of samples and pathology verify the success of emphysema model.2The effect of G-CSF in rabbit models ofemphysema:During the study,the Oxygen saturation of emphysema model grouphas a small fluctuation and was always lower than healthy controls.The heart rate of emphysema model group was always higher than healthy controls,B,C and D group has a small fluctuation,A group rised sharply in 6-8 week and then died.The oxygen saturationand heart rate between all groups and courses of emphysema model had no statistical significance(P>0.05).Inthefirst course,most emphysema models had a increased lung volume.In the second course,B group and C(b)increased a lot.In the third course,B(b)was decreased obviously and all groups changed almost nothing in the follow-up observation period,but it appeared to had tendency of reducement.MLI of A[(63.97±4.32)μm],B[(64.54±7.49)μm] and C group[(63.80±10.19)μm] were bigger than D group[(56.75±7.65)μm],and A,B were bigger than C group,but there was no statistical significance(P>0.05).PAP of each group had no statistically significant too(P>0.05).MLI of experimental groups(A,B and C)[(64.10±6.69)μm] was bigger than blank control(D)[(56.75±7.65)%],but there was no statistical significance(P>0.05).PAP of experimental groups(A,B and C)[(78.46±6.82)μm]and blank control(D)[(78.87±8.34)%]had no statistically significant too(P>0.05).Conclusions1The rabbit model of emphysema was successfully established.2Using G-CSF is a double-edged sword in the course of emphysema,it needs some conditions to play a role in the treatment of emphysema,or will aggravate it by oxidation and so on. |
