Objective:To observe the effect of g ADPN on the expression of TGF-1,Smad3,Smad7 and other related factors in the kidney of T2 DM rats,and to explore the protective mechanism of g ADPN on the occurrence and development of renal lesions in T2 DM rats.Methods:Male SD rats were randomly divided into normal control group(NC group),model group,.NC group was given normal diet.The model group was given high glucose and high fat diet.After 8 weeks,according to 30 mg / kg were given concentration of STZ 1%group were given intraperitoneal injection of.NC in lemon acid sodium citrate buffer at.4weeks after fasting blood glucose was higher than that of 16.7mmol / L,and the clinical manifestations of DM,showed that the T2 DM model has been successfully established.After 6 weeks of feeding to detection of urine protein of rats in model group were positive,the kidneys were observed,the model of type 2 diabetic nephropathy.After successful modeling,the model group were randomly divided into diabetic group(DM group),adiponectin group(AD group),AD group was given intraperitoneal injection of adiponectin ball type 10ug/kg,1 times a day,DM group received normal saline intraperitoneal injection 1 times a day,each group was fourth weeks,8 weeks,12 weeks at the end,collected blood samples,blood glucose;urine specimens,urine creatinine,and urine protein;kidney tissues were collected,weighed and measured the kidney weight index.To observe the pathological results,the determination of TGF-beta 1,Smad3,Smad7 m RNA and protein expression level.Results:1.Compared with normal control group and adiponectin group,the fasting blood glucose,24 h urinary albumin,urinary creatinine and kidney weight / body weight were significantly higher in the diabetic group(P<0.05);2.TGF-β1,Smad3 expression in the diabetic group were higher than the normal control group and the adiponectin group(P<0.05);3.Smad7 expression in the diabetic group were lower than the normal control group and the adiponectin group(P<0.05).Conclusion:1.TGF-β1,Smad3 play a pathogenic role in diabetic nephropathy,Smad7 play a protective role;2.Adiponectin can delay or even reverse the progression of diabetic nephropathy.
