Objective:The purpose of this study is to screen the serum microRNAs(miRNAs)which have different expressions between the chemoradiotherapy resistant patients and chemoradiotherapy sensitive patients of middle thoracic esophageal squamous cell carcinoma(ESCC)with stage Ⅱ-Ⅲ,to explore the relationship between serum miRNAs and chemoradiotherapy sensitivity of ESCC,and to find new biomarkers for predicting the efficacy of chemoradiotherapy for ESCC.Methods:55 ESCC patients with primary pathologically diagnosed were enrolled and treated with concurrent chemoradiotherapy.After the treatment the curative effect was evaluated according to the “evaluating standard of recent therapeutic efficacy for esophageal cancer after radiotherapy” of Wanjun in 2000 and response evaluation criteria in solid tumors(The version of RECIST1.1).And then all the patients were divided into chemoradiotherapy resistant group and chemoradiotherapy sensitive group.8 middle thoracic ESCC with stageⅡ-Ⅲ were selected from the two groups,whose clinical data had no statistical difference.The miScript miRNA PCR Array method was used to detect the expressions of 384 kinds of serum microRNA.And the serum miRNAs whose expressions were significantly different between the two groups were screened by the the criteria of fold change(FC)>2/<0.25 and t test P <0.05.Results:The expression of 8 kinds of serum miRNAs were significantly different between chemoradiotherapy resistant group and chemoradiotherapy sensitive group.The upregulated expressed miRNAs in the chemoradiotherapy resistance group were miR-21-5p(FC=9.6396,P=0.002),miR-223-3p(FC=5.4258,P=0.038),miR-423-5p(FC=6.2532,P= 0.025),miR-720(FC=8.1305,P=0.016),and the downregulated expressed miRNAs in the chemoradiotherapy resistance group were miR-16-5p(FC=0.1349,P=0.027),mi R-1280(FC=0.1554,P=0.041),miR-320a(FC=0.1419,P=0.032),miR-144-3p(FC=0.2038,P=0.049).Conclusion:1.This study successfully established a serum microRNA profile for middle thoracic ESCC patients with stage Ⅱ-Ⅲ.2.8 kinds of serum miRNAs were screened by this study: miR-21-5p,miR-423-5p,miR-720,miR-223-3p,miR-1280,miR-320 a,miR-144-3p,miR-16-5p.These miRNAs may be associated with the chemoradiotherapy sensitivity of middle thoracic ESCC with stageⅡ-Ⅲ,and could potentially become novel biomarkers predicting concurrent chemoradiotherapy therapeutic efficacy for middle thoracic ESCC with stage Ⅱ-Ⅲ.
