Effection Of Yishen Capsule On The Autophagy Of Mouse Podocytes Under High Glucose

Objective:Exploring the effection of Yishen Capsule on the autophagy in mouse podocytes under high glucose.Methods:With Yishen Capsule 0.625g/kg·d,1.25g/kg·d,2.5g/kg·d and benazepril 30mg/kg·d giving mice lavage for 7 days,then collecting blood sample from the abdominal aorta respectively to get different concentrations of medicated animal serum,which need centrifugation and sterilization.With the medicated serum culturing the mouse podocytes in vitro,the cells can be divided into following categories:normal group(NG),high glucose group(HG),Low concentration of Yishen Capsule group(LY),middle concentration of Yishen Capsule group(MY),high concentration of Yishen Capsule group(HY)and benazepril group(BP).Finally,detecting the expression changes of autophagy-related protein LC3Ⅱ and ubiquitin binding protein p62(p62 /SQSTMl)by Western blot,observing the autophagosome in each group by transmission electron microscope.Use the optical-microscope to observe the morphology of each group.Results:(1)The results of Western Blot: Compared with NG,the expression of autophagy-related protein LC3Ⅱ reduced in HG(p<0.05);Compared with HG,the expression of protein LC3Ⅱis increasing in all drug intervented groups,in which HY and BP increased significantly(p<0.05),whereas,the difference between HY and BPhave no statistical significance(p>0.05).Compared with NG,the expression of protein p62 increased in HG(p<0.05);Compared with HG,the expression of protein p62 reduced in all drug intervented groups,in which HY and BP reduced significantly(p<0.05),whereas,the difference between HY and BP have no statistical significance(p>0.05).(2)The results of transmission electron microscope:Compared with NG,the number of intracellular autophagosome decreased significantly in HG(p<0.05),Compared with HG,the the number of intracellular autophagosome is increasing in all drug intervented groups,in which HY and BP increased significantly(p<0.05),whereas,the difference between HY and BP have no statistical significance(p>0.05).(3)The results of optical-microscope:Cells,which rich in cell processes,is adherent to the Cell Culture Dish in NG.Compared with NG,there is much cells off from the Cell Culture Dish in HG.Compared with HG,cellular morphology is closer to NG in LY,MY,HY and BP,in which,HY and BP have more cell proliferation.Conclusion:High glucose inhibits the autophagy in podocytes,Yishen Capsule may have a certain improving effect on this autophagic inhibition to maintain a healthier kidney,of which the specific mechanism needs further research.
